      Optimasi Ekstraksi Senyawa Flavonoid Daun Miana (Coleus scutellarioides) dengan Desain I-optimal sebagai Antibakteri Penyebab Jerawat

      Abstract
      Daun miana (Coleus scutellarioides) mengandung flavonoid yang berpotensi sebagai agen antibakteri alami. Penelitian ini bertujuan mengoptimalkan ekstraksi flavonoid daun miana dan mengevaluasi aktivitas antibakterinya terhadap Cutibacterium acnes dan Staphylococcus epidermidis. Ekstraksi dilakukan dengan metode sonikasi, kadar total flavonoid ditentukan menggunakan metode kolorimetri AlCl3, dan aktivitas antibakteri diuji melalui metode difusi cakram, dan pengaruh konsentrasi ekstrak terhadap aktivitas antibakteri. Hasil verifikasi menunjukkan bahwa sampel ekstrak mampu menghambat pertumbuhan kedua bakteri dengan aktivitas tertinggi ditunjukkan sampel 2, dengan diameter zona hambat 9,03 ± 1,60 mm terhadap C. acnes dan 9,98 ± 0,40 mm terhadap S. epidermidis pada konsentrasi 100 mg/mL. Seluruh sampel bersifat bakteriostatik pada rentang konsentrasi yang diuji. Hasil penelitian menunjukkan bahwa ekstrak flavonoid daun miana efektif menghambat pertumbuhan bakteri penyebab jerawat.
       
      Miana leaves (Coleus scutellarioides) contain flavonoids with potential as natural antibacterial agents. This study aimed to optimize the extraction of flavonoids from miana leaves and evaluate their antibacterial activity against Cutibacterium acnes and Staphylococcus epidermidis. Extraction was carried out using the sonication method, the total flavonoid content was determined using the AlCl3 colorimetric method, and antibacterial activity was tested using the disc diffusion method, along with the effect of concentration on antibacterial activity. Verification results showed that the extract samples were able to inhibit the growth of both bacteria, with the highest activity shown by sample 2, with inhibition zone diameters of 9.03 ± 1.60 mm against C. acnes and 9.98 ± 0.40 mm against S. epidermidis at a concentration of 100 mg/mL. All samples exhibited bacteriostatic properties within the tested concentration range. The results indicate that the flavonoid extract from miana leaves is effective in inhibiting the growth of acnecausing bacteria.
