| dc.description.abstract | Daun sirih hijau (Piper betle L.) secara tradisional digunakan untuk menjaga kesehatan area kewanitaan, mengatasi infeksi dan iritasi kulit. Peningkatan resistensi terhadap antimikroba mendorong pencarian alternatif yang lebih efektif dan aman, salah satunya dengan mengeksplorasi cendawan endofit, yaitu mikroba yang hidup di dalam jaringan tanaman tanpa menimbulkan gejala pada inangnya. Penelitian ini bertujuan mengkaji potensi cendawan endofit asal daun sirih hijau (Piper betle L.) dalam menghambat bakteri patogen Staphylococcus aureus dan Escherichia coli, serta cendawan patogen Candida albicans. Isolasi cendawan endofit dilakukan dengan mendesinfeksi permukaan sampel daun, kemudian potongan jaringan daun diinokulasi pada media Potato Sucrose Agar (PSA) yang mengandung 0,05% kloramfenikol. Pengujian aktivitas antimikroba dilakukan menggunakan metode agar plug assay dengan modifikasi. Hasil penelitian diperoleh empat isolat cendawan endofit dengan karakteristik morfologi dan aktivitas antimikroba yang berbeda. Isolat SH4 menunjukkan aktivitas antimikroba terbaik karena mampu menghambat dua mikroba uji, yaitu S. aureus dengan nilai 2,51 mm dan C. albicans 2,17 mm. Cendawan endofit asal daun sirih hijau menunjukkan aktivitas antimikroba, namun potensi yang dihasilkan masih terbatas. | |
| dc.description.abstract | Green betel (Piper betle L.) leaves have traditionally been used to maintain
feminine hygiene and to treat infections and skin irritation. The rising prevalence
of antimicrobial resistance has driven the search for safer, more effective
alternatives, including the exploration of endophytic fungi, microorganisms that
inhabit plant tissues without causing symptoms in their host. This study aimed to
evaluate the potential of endophytic fungi isolated from green betel (Piper betle L.)
leaves to inhibit the pathogenic bacteria Staphylococcus aureus and Escherichia
coli, as well as the pathogenic fungus Candida albicans. Endophytic fungi were
isolated by surface sterilization of leaf samples, followed by the inoculation of leaf
segments onto potato sucrose agar (PSA) supplemented with 0,05%
chloramphenicol. Antimicrobial activity was evaluated using a modified agar plug
assay. Four endophytic fungal isolates with distinct morphological characteristics
and antimicrobial activities were obtained. Isolate SH4 exhibited the strongest
antimicrobial activity, inhibiting two test microorganisms, namely S. aureus with
an inhibition zone of 2.51 mm and C. albicans with a zone of 2.17 mm. While
endophytic fungi isolated from green betel leaves demonstrate clear antimicrobial
activity, their current potential remains limited. | |
