Propolis merupakan senyawa resin yang merupakan campuran dari air liur, lilin, dan hasil metabolisme cabang, bunga, serbuk sari, tunas, dan eksudat pohon pada lebah. Propolis sudah lama dikenal karena memiliki banyak manfaat bagi kesehatan. Propolis dan ekstrak propolis bersifat antiseptik, antiinflamasi, antioksidan, antibakteri, antijamur, antimaag, antikanker, dan imunomodulator. Komponen bioaktif propolis bervariasi tergantung pada sumber tanaman yang dihisap oleh lebah. Komponen yang umumnya terkandung pada propolis yakni lilin, resin, balsam, minyak atsiri, serta metabolit primer dan sekunder dari tanaman yang dihisap seperti asam amino, vitamin, fenol, terpenoid, dan alkaloid. Tujuan umum dari penelitian ini adalah untuk meneliti komposisi kimia propolis dari lebah Trigona laeviceps asal Jawa Barat dan mengidentifikasi komponen antibakteri dari propolis dari lebah Trigona laeviceps dengan pendekatan metabolomik.
Penelitian ini terdiri dari dua tahap. Tahap pertama terdiri dari karakterisasi propolis Trigona laeviceps mentah (analisis lilin, gum, proksimat, total fenol, tanin, flavonoid, alkaloid, dan saponin), ekstraksi propolis Trigona laeviceps dengan 3 pelarut, dan uji aktivitas antibakteri ekstrak propolis Trigona laeviceps dengan menggunakan metode difusi cakram. Pelarut yang digunakan yaitu akuades (polar), etanol (semi polar), dan n-heksanaa (non polar). Tahap kedua terdiri dari profiling senyawa pada ekstrak propolis Trigona laeviceps dari tiga wilayah di Jawa Barat yaitu Bandung, Sumedang, dan Purwakarta dengan pendekatan metabolomik; uji aktivitas antibakteri dari senyawa pada ekstrak propolis Trigona laeviceps dengan konsentrasi hambat minimum (KHM)/minimum inhibitory concentration (MIC). Data aktivitas antibakteri dianalisis dengan one way Analysis of Varian (ANOVA) pada perangkat Statistical Product and Service Solutions (IBM SPSS) versi 27 dan dilanjutkan dengan uji jarak berganda Duncan pada taraf 5%. Analisis data multivariat komponen aktif diterapkan dengan menggunakan area puncak senyawa serta mengorelasikan dengan aktivitas antibakteri dengan teknik Orthogonal Projection to Latent Structure (OPLS). Data OPLS dianalisis menggunakan software SIMCA versi 18.
Hasil penelitian tahap pertama menunjukkan komponen terbesar dari propolis mentah lebah Trigona laeviceps asal Bogor adalah lemak (65,47%). Berdasarkan hasil uji kualitatif, lilin, gum, tanin, flavonoid, dan alkaloid terdeteksi dalam propolis mentah. Kandungan fenol pada sampel sebesar 3,08 GAE/g, tanin sebesar 34,45 mg/g, dan flavonoid sebesar 21,88 mgQCE/g. Semua sampel tidak memiliki zona hambat terhadap bakteri Gram-negatif (Escherichia coli dan Salmonella Typhimurium). Sementara itu, pada bakteri Gram-positif (Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, dan Streptococcus mutans), ekstrak etanol propolis menunjukkan zona hambat tertinggi (16,92 mm). Hasil penelitian menunjukkan bahwa etanol merupakan pelarut yang paling sesuai untuk mengekstrak komponen antibakteri pada propolis dengan metode difusi cakram. Hasil penelitian ini dapat dimanfaatkan untuk meningkatkan nilai jual dari industri propolis lebah Trigona laevicep di Bogor.
Hasil penelitian tahap kedua menunjukkan bahwa ekstrak heksana memiliki aktivitas antibakteri tertinggi pada seluruh lokasi pengambilan sampel (p<0,05), dengan rentang nilai MIC antara 2,92% hingga 10,42%. Analisis antibakteri menggunakan metode MIC karena metode difusi cakram pada penelitian tahap pertama diduga tidak efektif untuk sampel ekstrak propolis dengan heksana. Analisis multivariat, meliputi principal component analysis (PCA) dan orthogonal partial least squares–discriminant analysis (OPLS–DA), menunjukkan bahwa variasi profil kimia ekstrak berasosiasi dengan polaritas pelarut yang digunakan. Analisis OPLS mengidentifikasi senyawa yang berkorelasi positif dengan aktivitas antibakteri terhadap Escherichia coli, Listeria monocytogenes, Staphylococcus aureus dan Salmonella Typhimurium yaitu asam oleat, ester geranil asetat, asam stearat, a-mangostin, dan kaempferol. Temuan ini mengindikasikan bahwa pelarut non-polar, khususnya heksana, lebih efektif dalam mengekstraksi komponen antibakteri dari propolis T. laeviceps, sehingga menunjukkan potensinya sebagai antibakteri alami. Propolis is a resinous compound composed of a mixture of saliva, wax, and the metabolic products of bees derived from branches, flowers, pollen, buds, and tree exudates. Propolis has long been recognized for its numerous health benefits. Both propolis and its extracts exhibit antiseptic, anti-inflammatory, antioxidant, antibacterial, antifungal, anti-ulcer, anticancer, and immunomodulatory properties. The bioactive components of propolis vary depending on the plant sources visited by the bees. Propolis generally contains wax, resin, balsam, essential oils, as well as primary and secondary plant metabolites such as amino acids, vitamins, phenolic compounds, terpenoids, and alkaloids. The general objective of this study was to investigate the chemical composition of propolis produced by Trigona laeviceps from West Java and to identify the antibacterial compounds in propolis from Trigona laeviceps using a metabolomic approach.
This study consisted of two stages. The first stage included the characterization of raw Trigona laeviceps propolis (wax, gum, proximate, total phenol, tannin, flavonoid, alkaloid, and saponin analysis), extraction of Trigona laeviceps propolis using three solvents, and antibacterial activity testing of the propolis extracts using the disc diffusion method. The solvents used were distilled water (polar), ethanol (semi-polar), and n-hexane (non-polar). The second stage involved compound profiling of Trigona laeviceps propolis extracts from three regions in West Java, namely Bandung, Sumedang, and Purwakarta, using a metabolomic approach; and antibacterial activity testing of the compounds in the propolis extracts using the minimum inhibitory concentration (MIC) method. Antibacterial activity data were analyzed using one-way Analysis of Variance (ANOVA) in the Statistical Product And Service Solutions (IBM SPSS) version 27, followed by Duncan’s multiple range test at a 5% significance level. Multivariate analysis of active compounds was conducted using compound peak areas and correlating them with antibacterial activity using the Orthogonal Projection to Latent Structure (OPLS) technique. OPLS data were analyzed using SIMCA software version 18.
The results of the first stage showed that the major component of raw Trigona laeviceps propolis from Bogor was fat (65.47%). Based on qualitative tests, wax, gum, tannins, flavonoids, and alkaloids were detected in the raw propolis. The phenolic content of the sample was 3.08 GAE/g, tannin content was 34.45 mg/g, and flavonoid content was 21.88 mgQCE/g. None of the samples exhibited inhibitory zones against Gram-negative bacteria (Escherichia coli and Salmonella Typhimurium). In contrast, against Gram-positive bacteria (Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, and Streptococcus mutans), the ethanol extract of propolis showed the highest inhibitory zone (16.92 mm). The results indicated that ethanol was the most suitable solvent for extracting antibacterial compounds from propolis using the disc diffusion method. These findings may contribute to increasing the commercial value of the Trigona laeviceps propolis industry in Bogor.
The results of the second stage demonstrated that the hexane extract exhibited the highest antibacterial activity across all sampling locations (p < 0.05), with MIC values ranging from 2.92% to 10.42%. Antibacterial analysis was conducted using the MIC method because the disc diffusion method applied in the preliminary stage was presumed ineffective for hexane propolis extracts. Multivariate analyses, including principal component analysis (PCA) and orthogonal partial least squares–discriminant analysis (OPLS–DA), revealed that variations in the chemical profiles of the extracts were associated with the polarity of the solvents used. OPLS analysis identified compounds that were positively correlated with antibacterial activity against Escherichia coli, Listeria monocytogenes, Staphylococcus aureus, and Salmonella Typhimurium, namely oleic acid, geranyl acetate, stearic acid, kaempferol, and a-mangostin. These findings indicate that non-polar solvents, particularly hexane, are more effective in extracting antibacterial compounds from Trigona laeviceps propolis, highlighting its potential as a natural antibacterial agent.
