      Produksi Eumelanin dari Isolat Bakteri Bacillus sp. CB 68 pada Beberapa Media Alternatif dan Evaluasi Potensinya Sebagai Antioksidan

      Abstract
      Melanin merupakan agen antioksidan alami potensial, namun bioproduksi massalnya terkendala oleh tingginya biaya prekursor murni. Penelitian ini bertujuan mengevaluasi produksi eumelanin oleh isolat CR 68 menggunakan media alternatif agroindustri, menguji kapasitas antioksidannya, dan mengonfirmasi identitas molekuler isolat. Produksi melanin dievaluasi pada media ekstrak kulit semangka, kulit melon, dan kedelai dibandingkan dengan media kontrol L-tirosin. Karakterisasi tipe pigmen dilakukan melalui spektrofotometri UV-Vis, sedangkan aktivitas antioksidan diuji menggunakan metode penangkapan radikal DPPH. Identifikasi taksonomi isolat dianalisis melalui amplifikasi gen 16S rRNA. Hasil menunjukkan perolehan rendemen melanin tertinggi terdapat pada media L-tirosin (234 mg/L) dan ekstrak kedelai (166 mg/L). Nilai kesetaraan kemurnian melanin paling tinggi dicapai oleh media ekstrak kulit semangka (148,08 µg/mg). Seluruh ekstrak pigmen terkonfirmasi sebagai eumelanin berdasarkan rasio absorbansi A650/A500 > 0,25. Uji DPPH menunjukkan ekstrak eumelanin tergolong antioksidan lemah, dengan nilai IC50 terbaik ditunjukkan oleh medium kedelai (747,43 µg/mL). Analisis filogenetik mengonfirmasi isolat CR 68 sebagai spesies Bacillus cereus. Pemanfaatan limbah agroindustri, khususnya kulit semangka, memiliki potensi sebagai substrat ekonomis dalam efisiensi bioproduksi eumelanin.
       
      Melanin is a potential natural antioxidant, but its large-scale bioproduction is limited by high precursor costs. This study aimed to evaluate eumelanin production by isolate CR 68 using alternative agro-industrial media, assess its antioxidant capacity, and confirm the isolate's molecular identity. Pigment production was evaluated in watermelon rind, melon rind, and soybean extract media compared to a standard L-tyrosine medium. Pigment characterization was performed using UV Vis spectrophotometry, while antioxidant activity was tested using the DPPH radical scavenging assay. Taxonomic identification was conducted through 16S rRNA gene amplification. The highest melanin yield was obtained in L-tyrosine (234 mg/L) and soybean extract (166 mg/L) media. Nevertheless, the highest equivalent melanin purity was achieved in watermelon rind extract (148.08 µg/mg). All extracted pigments were confirmed as eumelanin based on an A650/A500 absorbance ratio > 0.25. The DPPH assay revealed that the extracts exhibited weak antioxidant activity, with the best IC50 value derived from the soybean medium (747.43 µg/mL). Phylogenetic analysis definitively identified isolate CR 68 as Bacillus cereus. Agro-industrial wastes, particularly watermelon rind, hold significant potential as economical substrates for efficient eumelanin bioproduction.
