      Optimasi Suhu Dan Lama Penyeduhan Teh Celup Komersil Untuk Ekstraksi Senyawa Bioaktif

      Abstract
      Black tea is one of the most widely consumed beverages in Indonesia and contains bioactive compounds that contribute to antioxidant activity. Brewing temperature and time influence the extraction efficiency of these compounds, making the determination of optimal brewing conditions essential. This study aimed to optimize the brewing temperature (62, 77, and 85 °C) and brewing time (15, 120, and 300 s) of commercial tea bags. Analyses included total phenolic content (TPC), antioxidant activity using DPPH and FRAP assays, followed by two-way ANOVA and Tukey's HSD test (p<0.05). The highest TPC was obtained at 77 °C for 300 s (156.699 ± 8.980 mg GAE/100 mL), whereas the highest FRAP antioxidant activity was observed at 85 °C for 300 s (513.328 ± 9.899 mg AAE/100 mL). The DPPH assay showed no significant differences among most treatments. Brewing at 85 °C for 300 s is recommended as the optimal condition for extracting bioactive compounds.
       
      Teh hitam merupakan salah satu minuman yang paling banyak dikonsumsi di Indonesia dan mengandung senyawa bioaktif yang berkontribusi terhadap aktivitas antioksidan. Suhu dan lama penyeduhan memengaruhi efisiensi ekstraksi senyawa tersebut sehingga diperlukan kondisi penyeduhan yang optimal. Penelitian ini bertujuan mengoptimasi suhu (62, 77, dan 85 °C) dan lama penyeduhan (15, 120, dan 300 detik) teh celup komersial. Analisis meliputi kadar total fenolik (TPC), aktivitas antioksidan metode DPPH dan FRAP, serta analisis statistik menggunakan two-way ANOVA dan uji Tukey HSD (p<0,05). TPC tertinggi diperoleh pada suhu 77 °C selama 300 detik sebesar 156,699 ± 8,980 mg GAE/100 mL, sedangkan aktivitas antioksidan FRAP tertinggi diperoleh pada suhu 85 °C selama 300 detik sebesar 513,328 ± 9,899 mg AAE/100 mL. Aktivitas antioksidan metode DPPH tidak menunjukkan perbedaan nyata pada sebagian besar perlakuan. Suhu 85 °C selama 300 detik direkomendasikan sebagai kondisi penyeduhan optimal untuk mengekstrak senyawa bioaktif.
