      Abstract
      Fermentasi pada tanaman obat merupakan salah satu pendekatan untuk meningkatkan konsentrasi dan bioaktivitas senyawa metabolit. Bangle (Zingiber montanum) diketahui mengandung berbagai senyawa kimia dengan beragam khasiat farmakologis, termasuk antikanker. Penelitian ini bertujuan mengidentifikasi profil senyawa ekstrak bangle fermentasi dan nonfermentasi serta mengevaluasi potensinya dalam menginduksi apoptosis pada sel ragi. Fermentasi padat dilakukan menggunakan Aspergillus oryzae selama 14 hari, kemudian profil senyawa kimia diidentifikasi menggunakan LC-MS/MS. Kemampuan induksi apoptosis dievaluasi melalui perhitungan koloni petite dan uji aktivitas mitokondria menggunakan pewarnaan Rhodamin B serta Annexin V/FITC-PI. Hasil penelitian menunjukkan bahwa fermentasi menyebabkan perubahan kelimpahan relatif metabolit ekstrak bangle. Selain itu, ekstrak fermentasi 14 hari meningkatkan frekuensi koloni petite dan mengganggu fungsi mitokondria. Oleh karena itu, dapat disimpulkan bahwa fermentasi padat bangle dengan A. oryzae mengubah profil metabolit dan meningkatkan potensi induksi apoptosis.
       
      Fermentation of medicinal plants is one approach used to enhance the concentration and bioactivity of secondary metabolites. Bangle (Zingiber montanum) is known to contain various bioactive compounds with diverse pharmacological properties, including anticancer potential. This study aimed to identify the metabolite profiles of fermented and nonfermented bangle extracts and to evaluate their potential to induce apoptosis in yeast cells. Solid-state fermentation was carried out using Aspergillus oryzae for 14 days, followed by metabolite profiling using LC-MS/MS. Apoptosis-inducing activity was assessed through petite colony formation analysis, mitochondrial activity evaluation using Rhodamine B staining, and Annexin V/FITC-PI staining. The results showed that fermentation altered the relative abundance of metabolites in bangle extracts. Furthermore, the 14-day fermented extract increased the frequency of petite colonies and disrupted mitochondrial function. These findings indicate that solid-state fermentation of bangle with A. oryzae modifies the metabolite profile and enhances its apoptosis-inducing potential.
