dc.description.abstractKelapa sawit merupakan komoditas perkebunan strategis yang produktivitasnya sangat dipengaruhi oleh kualitas bibit. Salah satu gangguan yang berpotensi menurunkan kesehatan bibit ialah nematoda parasit tumbuhan yang menyerang sistem perakaran. Penelitian ini bertujuan mengidentifikasi nematoda yang berasosiasi dengan bibit kelapa sawit pada tahap pre-nursery dan main- nursery di PT Perkebunan Nusantara IV Regional 2, Sumatra Utara. Pengambilan sampel dilakukan secara purposif pada bibit bergejala, masing-masing sebanyak 10 polibag pre-nursery berumur 10 minggu dan 10 polibag main-nursery berumur 36 minggu. Sampel tanaman kelapa sawit liar dari Kebun Percobaan Cikabayan IPB digunakan sebagai pembanding. Nematoda diekstraksi dari akar menggunakan metode mist chamber dan dari tanah menggunakan metode flotasi- sentrifugasi. Identifikasi dilakukan berdasarkan karakter morfologi dan morfometri menggunakan kunci identifikasi serta formula De Man. Hasil penelitian menunjukkan dua genus fitonematoda, yaitu Rotylenchulus di pre- nursery Lokasi 3 dan main-nursery Lokasi 2 dan Helicotylenchus di pre-nursery Lokasi 3, serta lima genus nematoda non-parasit, yaitu Alaimus, Dorylaimus, Cephalobus, Mononchus, dan Rhabditis. Rotylenchulus ditemukan pada akar dan tanah main-nursery serta sampel referensi, sedangkan Helicotylenchus hanya ditemukan pada tanah sampel referensi. Nematoda non-parasit memiliki kelimpahan dan keragaman lebih tinggi dibandingkan fitonematoda, terutama pada lokasi main-nursery. Lokasi pre-nursery menunjukkan jumlah nematoda paling rendah. Gejala bibit meliputi pertumbuhan terhambat, kerdil, daun menguning, memerah, melintir, berkerut, dan pertumbuhan tidak merata selama kegiatan survei.
dc.description.sponsorshipOil palm is a strategic plantation commodity whose productivity is strongly influenced by seedling quality. One factor that may reduce seedling health is plant-parasitic nematodes that attack the root system. This study aimed to identify nematodes associated with oil palm seedlings at the pre-nursery and main-nursery stages at PT Perkebunan Nusantara IV Regional 2, North Sumatra. Sampling was conducted purposively on symptomatic seedlings, consisting of 10 pre-nursery polybags at 10 weeks of age and 10 main-nursery polybags at 36 weeks of age. Wild oil palm samples from the Cikabayan Experimental Garden, IPB, were used as a reference. Nematodes were extracted from roots using the mist chamber method and from soil using the flotation-centrifugation method. Identification was carried out based on morphological and morphometric characters using identification keys and the De Man formula. The results showed two genera of plant-parasitic nematodes, namely Rotylenchulus (found in pre- nursery Location 3 and main-nursery Location 2) and Helicotylenchus (found in pre-nursery Location 3), and five genera of non-parasitic nematodes, namely Alaimus, Dorylaimus, Cephalobus, Mononchus, and Rhabditis. Rotylenchulus was found in the roots and soil of the main-nursery samples and in the reference sample, whereas Helicotylenchus was found only in the soil of the reference sample. Non-parasitic nematodes showed higher abundance and diversity than plant-parasitic nematodes, particularly in the main-nursery location. The pre- nursery location showed the lowest number of nematodes. Seedling symptoms included stunted growth, dwarfing, yellowing, reddening, leaf curling, wrinkling, and uneven growth during the survey.
