      Bacillus sp. sebagai Bakteri Pemacu Tumbuh Padi In Vitro

      Abstract
      Rizobakteri yang mampu memacu pertumbuhan tanaman dikenal sebagai Plant Growth-Promoting Rhizobacteria (PGPR), yang berpotensi menjadi solusi untuk mengurangi ketergantungan pada pupuk kimia. Salah satu kelompok PGPR yang potensial adalah genus Bacillus yang diisolasi dari perakaran tanaman padi. Penelitian ini bertujuan mengeksplorasi kemampuan Bacillus sp. dalam memacu pertumbuhan padi secara in vitro. Uji dilakukan pada produksi IAA, pemacu pertumbuhan, dan identifikasi isolat Bacillus potensial menggunakan gen 16S rRNA. Aktivitas pemacu tumbuh diuji berdasarkan 2 parameter yang menghasilkan 2 isolat signifikan. Hasil penelitian menunjukan 2 dari 6 isolat Bacillus yang diuji dengan kode N.1.01 dan N.1.05 memiliki potensi yang baik dalam memacu pertumbuhan padi. Isolat dengan kode N.1.01 memproduksi IAA sebesar 3,2 ppm dan N.1.05 memproduksi IAA sebesar 3,4 ppm. Kedua isolat memiliki produksi IAA yang lebih rendah, tetapi memiliki aktivitas pemacu tumbuh yang lebih tinggi. Analisis gen 16S rRNA kedua isolat menunjukan kemiripan tertinggi dengan Bacillus clarus strain ATCC 21929 dan Bacillus cereus ATCC 14579. Kedua bakteri teridentifikasi sebagai rizobakteri pemacu pertumbuhan tanaman (PGPR).
       
      Rhizobacteria capable of stimulating plant growth are known as Plant Growth-Promoting Rhizobacteria (PGPR), which have the potential to reduce dependence on chemical fertilizers. One promising group of PGPR is the genus Bacillus, isolated from rice plant roots. This study aimed to explore the ability of Bacillus sp. to stimulate rice growth in vitro. Tests were conducted on IAA production, a growth promoter, and the identification of potential Bacillus isolates using the 16S rRNA gene. Growth-promoting activity was assessed based on two parameters, yielding two significant isolates. The results showed that two of the six Bacillus isolates tested, designated N.1.01 and N.1.05, demonstrated good potential for promoting rice growth. The isolate coded N.1.01 produced 3,2 ppm of IAA, while N.1.05 produced 3,4 ppm of IAA. Both isolates had lower IAA production but exhibited higher growth-promoting activity. Analysis of the 16S rRNA genes of both isolates showed the highest similarity to Bacillus clarus strain ATCC 21929 and Bacillus cereus ATCC 14579. Both bacteria were identified as plant growth-promoting rhizobacteria (PGPR).
