Pentaploidisasi melalui Kejutan Suhu Panas pada Ikan Patin (Pangasianodon hypophthalmus)

Ikan patin (Pangasianodon hypophthalmus) menjadi pilihan alternatif sebagai sumber nutrien secara global, peralihan dari konsumsi ikan cod yang mulai dibatasi penangkapannya. Kondisi ini mendorong permintaan ikan patin di pasar dunia meningkat 25,4% pada tahun 2023-2025. Namun, untuk memenuhi permintaan yang terus meningkat, durasi pemeliharaannya relatif lambat akibat pertumbuhannya yang relatif rendah. Sehingga diperlukan upaya mempersingkat durasi pemeliharaan dengan rekayasa peningkatan pertumbuhan. Salah satu teknologi perbaikan mutu genetik yang dapat meningkatkan kecepatan tumbuh adalah rekayasa set kromosom. Poliploidisasi merupakan penambahan set kromosom lebih dari normal (2n), yang mampu menghasilkan benih unggul. Individu dengan set kromosom ganjil seperti triploid (3n) memiliki keunggulan pertumbuhan yang cepat karena bersifat steril dan tidak mengalami perkembangan gonad, sehingga energi untuk perkembangan gonad dialihkan ke pertumbuhan somatik. Produksi ikan triploid dapat dilakukan melalui kejut suhu panas, suhu dingin, kimia, tekanan, dan listrik. Metode kejut suhu panas sering diterapkan karena mudah dan aman terhadap lingkungan. Induksi ikan patin triploid melalui teknik kejut suhu panas pernah dilakukan dengan tingkat keberhasilan 70%. Produksi ikan patin triploid secara massal juga pernah dilakukan dengan tingkat keberhasilan 100%. Teknik rekayasa ini membuka peluang eksplorasi tingkat ploidi ganjil yang lebih tinggi seperti pentaploid (5n) pada ikan patin, sehingga perlu optimasi induksi pentaploid melalui kejut suhu panas dengan intensitas suhu dan umur zigot.
Penelitian ini menggunakan rancangan acak lengkap (RAL) faktorial dengan tiga taraf suhu (41, 42, 43ºC) dan tiga taraf umur zigot (1,5, 2, 2,5 menit setelah fertilisasi, msf), masing-masing dengan tiga kali ulangan. Pentaploidisasi dilakukan dari perkawinan induk betina tetraploid (4n) dan jantan diploid melalui paparan kejut suhu panas, sedangkan kelompok kontrol menggunakan induk betina dan jantan diploid, yang sebelumnya dilakukan penyuntikan hormon Human Chorionic Gonadotropin pada betina dan ovaprim pada betina serta jantan. Telur dan sperma dicampur secara buatan, setelah diaktivasi dan umur zigot mencapai masing-masing perlakuan, dilakukan paparan kejut suhu panas dengan masing-masing intensitas sesuai perlakuan. Telur hasil kejutan dipindahkan pada wadah penetasan untuk diamati derajat pembuahan, penetasan dan abnormalitas. Larva yang menetas dipindahkan pada media pemeliharaan dengan pemberian pakan artemia pada minggu pertama dan cacing sutera pada minggu kedua. Pada akhir pemeliharaan, diamati derajat kelangsungan hidup dan keberhasilan pentaploid melalui perhitungan jumlah nukleolus dan dikonfirmasi dengan analisis jumlah kromosom menggunakan teknik kultur darah. Uji histologi gonad dilakukan terhadap individu yang terkonfirmasi pentaploid, triploid, dan diploid pada umur 14 bulan.
Derajat pembuahan menunjukkan nilai tertinggi pada perlakuan kombinasi suhu 43°C dengan umur zigot 2,5 msf (57,22 ± 9,11%) dan derajat penetasan menunjukkan nilai tertinggi pada perlakuan kontrol (42,96 ± 10,25%), yang keduanya berbeda signifikan terhadap beberapa perlakuan (p<0,05). Derajat pembuahan dan penetasan menunjukkan tren penurunan pada umur zigot yang singkat. Menariknya, suhu 43°C dapat mengurangi dampak negatif tersebut, diduga ikan patin mengalami kematangan fisiologis yang lebih lama dan toleransi suhu tinggi yang mampu meningkatkan laju metabolisme embrio. Pada derajat abnormalitas, perlakuan suhu 43°C pada 2,5 msf menunjukkan nilai tertinggi (24,04 ± 7,75%), sedangkan kelompok kontrol menghasilkan nilai terendah, artinya paparan kejut suhu panas mengakibatkan peningkatan risiko deformasi morfologis pada larva. Kondisi ini mengindikasikan adanya trade-off yang tidak terhindarkan antara keberhasilan induksi kromosom dan integritas perkembangan embrio. Derajat kelangsungan hidup menunjukkan nilai tertinggi pada perlakuan suhu 41°C pada 1,5 msf (43,20 ± 3,65%), yang berbeda signifikan terhadap kelompok kontrol dan beberapa perlakuan (p<0,05). Diduga peningkatan karakter kanibalisme akibat kepadatan tinggi menjadi faktor utama mempengaruhi tingkat kelangsungan hidup.
Faktor suhu memberikan perbedaan signifikan terhadap keberhasilan induksi (p<0,05), dan perlakuan suhu 43°C menunjukkan keberhasilan induksi pentaploid tertinggi (20,63 ± 4,83 %). Hasil plot interaksi menunjukkan kombinasi suhu 43°C dengan umur zigot 2 msf menghasilkan nilai keberhasilan induksi tertinggi secara numerik. Induk ikan patin betina tetraploid yang digunakan diduga membutuhkan intensitas kejutan lebih tinggi (43°C) untuk menghambat polar body kedua selama meiosis II. Keberhasilan induksi menghasilkan individu pentaploid dengan jumlah maksimum 5 nukleolus per sel dan 135 kromosom, menandakan terhambatnya pelepasan polar body kedua pada telur 4n, kemudian berfusi dengan gamet haploid (n) dari sperma. Sementara itu, kegagalan induksi menghasilkan individu triploid dengan jumlah maksimum 3 nukleolus per sel dan 81 kromosom. Pada uji histologi, tidak ditemukan sel germinal yang berkembang pada ovari ikan pentaploid, sedangkan perkembangan testis ikan pentaploid terhenti sampai tahap spermatogonia. Kerusakan gonad pentaploid diduga diakibatkan kegagalan saat proses sinapsis, yang selanjutnya berdampak pada gangguan gametogenesis dan mengarah pada indikasi sterilitas.
Penelitian ini menyimpulkan bahwa keberhasilan ikan patin pentaploid terbaik diperoleh melalui kombinasi kejut suhu panas 43ºC pada umur zigot 2 menit setelah fertilisasi, dengan durasi 2 menit. Jumlah kromosom ikan patin pentaploid yang dihasilkan adalah 5n = 135. Serta, kondisi gonad ikan patin pentaploid yang tidak berkembang mengindikasikan sterilitas. Perlu mengevaluasi lebih lanjut terkait karakter pertumbuhan dan metode produksi massal ikan patin pentaploid (5n) yang optimal. Striped catfish (Pangasianodon hypophthalmus) has emerged as an alternative global source of animal protein, partly due to restrictions on cod fisheries. This condition drove a 25,4% increase in global demand for striped catfish between 2023-2025. However, the increasing demand is constrained by relatively slow growth and extended culture duration. Therefore, strategies to accelerate growth performance are required. Chromosome set manipulation is a promosing approach. Polyploidization, defined as the addition of chromosome sets beyond the normal diploid (2n) condition, has been widely applied to produce superior fish strains. Triploid fish (3n), possessing an odd number of chromosome sets, generally exhibit enhanced growth due to gonadal sterility, whereby energy allocation is shifted from reproduction to somatic growth. Triploid production can be induced through heat shock, cold shock, chemical agents, pressure, or electrical stimulation, with heat shock being widely preferred due to its simplicity, cost effectiveness, and environmental safety. While triploidization in striped catfish has been successfully achieved, the production of pentaploid (5n) individuals using heat shock has not yet been reported. Therefore, this study aimed to explore the induction of pentaploid using heat shock at different temperature intensities and initial times.
This study employed a completely randomized factorial design consisting of three heat shock temperatures (41, 42, and 43°C) and three initial times (1,5, 2,0, and 2,5 min post fertilization, mpf), with three replicates for each treatment combination. Pentaploidization was induced by crossing tetraploid (4n) females with diploid males, followed by heat shock treatment, whereas the control group consisted of diploid females crossed with diploid males. Prior to artificial fertilization, females were injected with human chorionic gonadotropin (hCG) followed by Ovaprim, while males received Ovaprim only. Eggs and sperm were manually stripped and fertilized, and after activation, zygotes were exposed to heat shock at the designated developmental age according to each treatment. Heat-shocked eggs were subsequently transferred to hatching containers for the assessment of fertilization rate, hatching rate, and larval abnormalities. Hatched larvae were reared in culture containers and fed Artemia nauplii during the first week, followed by Tubifex worms during the second week. At the end of the rearing period, survival rate and pentaploid induction efficiency were evaluated based on nucleolar counts and subsequently confirmed by chromosome analysis using a blood culture technique. Gonadal histological analysis was performed on confirmed pentaploid, triploid, and diploid individuals at 14 months of age.
The highest fertilization rate was observed in the treatment combining a heat shock temperature of 43°C with a initial time of 2,5 mpf (57,22 ± 9,11%), whereas the highest hatching rate was recorded in the control group (42,96 ± 10,25%). Both values differed significantly from those of several other treatments (p<0,05). Fertilization and hatching rates exhibited a declining trend at earlier initial times. Interestingly, the adverse effects associated with shorter initial times were partially alleviated by the 43°C heat shock treatment. This response may be attributed to the relatively prolonged physiological maturation of striped catfish embryos and their tolerance to elevated temperatures, which may enhance embryonic metabolic activity. The highest incidence of larval abnormalities was recorded in the treatment exposed to 43°C at 2,5 mpf (24,04 ± 7,75%), whereas the control group exhibited the lowest incidence. These findings indicate that heat shock treatment increases the risk of morphological deformities in larvae. This outcome suggests an unavoidable trade-off between successful chromosome-set manipulation and the maintenance of normal embryonic development. The highest survival rate was observed in the treatment exposed to 41°C at 1,5 mpf (43,20 ± 3,65%), which differed significantly from the control group and several other treatments (p<0,05). The relatively lower survival observed in the remaining treatments was likely associated with increased cannibalistic behavior resulting from higher stocking density, which is presumed to be the primary factor influencing larval survival.
Temperature had a significant effect on pentaploid induction efficiency (p<0,05), with the 43°C treatment producing the highest induction rate (20,63 ± 4,83%). Interaction plot analysis further indicated that the combination of a 43°C heat shock treatment and a initial time of 2,0 mpf yielded the highest numerical induction efficiency. These findings suggest that tetraploid female striped catfish require a higher heat shock intensity (43°C) to effectively inhibit extrusion of the second polar body during meiosis II. Successful induction produced pentaploid individuals characterized by a maximum of five nucleoli per nucleus and a chromosome number of 135, confirming retention of the second polar body in tetraploid (4n) oocytes followed by fusion with a haploid (n) spermatozoon. In contrast, unsuccessful induction resulted in triploid individuals with a maximum of three nucleoli per nucleus and 81 chromosomes. Histological examination revealed no developing germ cells in the ovaries of pentaploid fish, whereas testicular development was arrested at the spermatogonial stage. The gonadal abnormalities observed in pentaploid individuals are likely attributable to impaired homologous chromosome pairing during meiosis, resulting in disrupted gametogenesis and indicating functional sterility.
This study demonstrated that the highest pentaploid induction efficiency in striped catfish was achieved by applying a heat shock treatment at 43°C for 2 min to zygotes at 2 mpf. The resulting pentaploid individuals possessed a chromosome complement of 5n = 135. Furthermore, the absence of gonadal development in pentaploid fish indicates functional sterility. Future studies should focus on evaluating the growth performance of pentaploid striped catfish and optimizing protocols for their large scale production.
