      Eksistensi dan Karakter Aktinobakteri Simbion Lebah Apis cerana dan Tetragonula laeviceps

      Abstract
      Aktinobakteri merupakan salah satu bakteri yang berhasil diisolasi pada saluran pencernaan lebah. Lebah pekerja Apis cerana dan Tetragonula laeviceps merupakan lebah penghasil madu yang dibudidayakan di Indonesia, namun eksplorasi aktinobakteri pada saluran pencernaan kedua jenis lebah ini belum banyak dieksplorasi. Penelitian ini bertujuan mengisolasi, mengkarakterisasi dan mengidentifikasi aktinobakteri di saluran pencernaan lebah pekerja A. cerana dan T. laeviceps berdasarkan gen 16S rRNA. Metode isolasi aktinobakteri saluran pencernaan lebah pekerja A. cerana dan T. laeviceps menggunakan metode pengayaan media selektif dan antibiotik. Aktinobakteri dikarakterisasi berdasarkan morfologi makroskopis pada tiga media pertumbuhan dan morfologi mikroskopis dengan metode slide culture. Isolat aktinobakteri diidentifikasi berdasarkan gen 16S rRNA. Aktinobakteri berhasil diisolasi dari saluran pencernaan lebah T. laeviceps sebanyak dua isolat yaitu isolat Tl1 dan Tl2, sedangkan aktinobakteri pada saluran pencernaan A. cerana belum dapat diisolasi dengan metode kultur sehingga diperlukan pendekatan metode lain. Kedua isolat aktinobakteri dari saluran pencernaan lebah T. laeviceps memiliki karakteristik morfologi warna miselieum substrat, aerial dan tesktur permukaan yang berbeda pada media pertumbuhan ISP-2, ISP-3, dan ISP-4. Tipe rantai spora pada kedua isolat yaitu fragmenting dan rectiflexibiles. Isolat Tl1 diduga termasuk aktinobakteri non-Streptomyces, sedangkan hasil identifikasi berdasarkan gen 16S rRNA isolat Tl2 memiliki kemiripan 99.27% dengan Streptomyces sampsonii strain ATCC 25495.
       
      Actinobacteria are among the bacterial groups that has been successfully isolated from the digestive tracts of bees. Beekeeping in Indonesia are widely use honey bees Apis cerana and stingless bees Tetragonula laeviceps. However, exploration of actinobacteria associated with their digestive tracts remains limited. This study aimed to isolate, characterize, and identify actinobacteria associated with the digestive tracts of worker bees A. cerana and T. laeviceps using 16S rRNA gene analysis. Actinobacteria were isolated using an enrichment method with selective media and antibiotic. Characterization actinobacteria by macroscopic morphology on three media and microscopic morphology using slide culture method. The isolates were identified based on the 16S rRNA gene. Two actinobacterial isolates, i.e. Tl1 and Tl2, were successfully isolated from the digestive tract of T. laeviceps. However, actinobacteria from the digestive tracts of A. cerana have not yet been success using culture-based methods, suggesting that alternative approaches are required to investigate the presence of actinobacteria. The isolates exhibited distinct substrate mycelia color, aerial mycelia, and colony surface textures on ISP-2, ISP-3, and ISP-4 media. Their spore chains were fragmenting and rectiflexibiles. Isolate Tl1 was predicted belong to a non-Streptomyces actinobacterium, whereas Tl2 showed 99.27% sequence similarity to Streptomyces sampsonii strain ATCC 25495.
