      Isolasi dan Karakteristik Bakteri dari Sawi Asin

      Abstract
      Fermentasi merupakan metode pengawetan makanan secara tradisional yang dilakukan secara turun-temurun. Salah satu produk pangan hasil fermentasi adalah sawi asin yang dibuat melalui proses perendaman dalam larutan garam tanpa penambahan kultur starter. Kondisi fermentasi tersebut mendukung pertumbuhan kelompok mikroorganisme tertentu, terutama bakteri asam laktat dan bakteri halofilik yang mampu bertahan pada kondisi salinitas tinggi. Penelitian ini bertujuan mengisolasi dan mengkarakterisasi bakteri dari sawi asin berdasarkan karakteristik morfologinya, serta melakukan skrining isolat bakteri melalui uji aktivitas hemolitik. Selain itu, penelitian ini juga bertujuan mengidentifikasi isolat bakteri terpilih secara molekuler menggunakan analisis gen 16S rRNA serta mengetahui kemampuan toleransi terhadap garam (NaCl) dan kemampuan produksi asam isolat terpilih. Satu isolat bakteri potensial (SA7) bersifat Gram positif dengan sel berbentuk batang berhasil diisolasi menggunakan media MRS padat +0,5% CaCO3+2% NaCl. Uji hemolisis pada agar-agar darah mengonfirmasi sifat non-hemolisis. Pewarnaan endospora menunjukkan bakteri tidak memiliki kemampuan membentuk endospora. Hasil identifikasi molekuler menyatakan bahwa isolat SA7 memiliki kemiripan sebesar 98,64% dengan Limosilactobacillus fermentum strain NBRC 15885 dengan nilai bootstrap 100. Isolat SA7 memiliki kemampuan toleransi terhadap kadar garam (NaCl) hingga 6% dan aktivitas produksi asam tertinggi pada hari ke-4 senilai 1,614% dan termasuk bakteri heterofermentatif obligat.
       
      Fermentation is a traditional food preservation method that has been practiced for generations. Pickled mustard greens are produced by fermenting the greens in a salt solution without the addition of a starter culture. The high salinity of the fermentation environment promotes the growth of specific microbial populations, particularly halophilic species and lactic acid bacteria. This study aimed to isolate and characterize bacteria from pickled mustard greens based on their morphological characteristics, screen the bacterial isolates for hemolytic activity, identify selected bacterial isolates molecularly using 16S rRNA gene analysis, and determine their salt (NaCl) tolerance and acid production capabilities. A potential Gram-positive bacterial isolate (SA7) with rod-shaped cells was successfully isolated using solid MRS medium supplemented with 0,5% CaCO3 and 2% NaCl. A hemolysis test on blood agar confirmed that the isolate was non-hemolysis. Endospore staining indicated that the bacterium is incapable of forming endospores. Molecular identification revealed that isolate SA7 shared 98,64% sequence similarity with Limosilactobacillus fermentum strain NBRC 15885, with a bootstrap value of 100. Isolate SA7 exhibits salt (NaCl) tolerance up to 6% and reached peak acid production on day 4 with a total acidity 1,614%. Based on its physiological characteristic, isolate SA7 was classified as an obligately heterofermentative bacterium.
