| dc.description.abstract | Kentang kultivar IPB CP1 dan CP4 merupakan kultivar lokal yang dikembangkan oleh Pusat Bioteknologi IPB untuk bahan baku industri kentang olahan. Sifat kultivar IPB CP1 dan CP4 masih perlu diperbaiki khususnya dalam hal ketahanannya terhadap penyakit. Upaya mutasi kultivar IPB CP1 dan CP4 telah dilakukan menggunakan iradiasi sinar gamma dosis 20, 30, 40, dan 60 Gy. Keberhasilan mutasi tersebut perlu dianalisis secara molekular untuk memastikan adanya perubahan genetik. Penelitian ini bertujuan menganalisis keragaman genetik klon kentang mutan dari kultivar IPB CP1 dan CP4 berdasarkan marka random amplified polymorphism DNA (RAPD) dan inter simple sequence repeat (ISSR). DNA genom diisolasi menggunakan metode cetyl trimethyl ammonium bromide (CTAB), lalu diamplifikasi dengan sepuluh primer RAPD dan delapan primer ISSR. Hasil amplifikasi menunjukkan polimorfisme sebesar 46,28% untuk RAPD dan 44,43% untuk primer ISSR. Temuan ini mengindikasikan bahwa mutasi planlet kentang dapat diinduksi menggunakan sinar gamma hingga dosis 20 Gy. Marka molekuler RAPD dan ISSR mampu menunjukkan adanya keragaman genetik di antara klon–klon mutan hasil iradiasi sinar gamma. | |
| dc.description.abstract | IPB CP1 and CP4 potato cultivars are local varieties developed by IPB Biotechnology Center as raw materials for processed potato industry. Characteristics of these cultivars still need improvement, particularly in disease resistance. Mutation induction in IPB CP1 and CP4 cultivars have been conducted using gamma ray irradiation at doses 20, 30, 40, and 60 Gy. The success of these mutations needs to be analyzed molecularly to confirm presence of genetic changes. This study aims to analyze genetic diversity of mutant potato clones from IPB CP1 and CP4 cultivars based on random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR). Genomic DNA was isolated using cetyl trimethyl ammonium bromide (CTAB) method, then amplified with ten RAPD primers and eight ISSR primers. The amplification results showed polymorphism rate of 46.28% for RAPD and 44.43% for ISSR. These findings indicate that mutations in potato plantlets can be induced using gamma rays up to a dose of 20 Gy. RAPD and ISSR were able to demonstrate presence of genetic diversity among mutant clones resulting from gamma ray irradiation. | |
