| dc.description.abstract | Staphylococcus aureus merupakan bakteri yang dapat membentuk biofilm
sehingga infeksi menjadi lebih sulit ditangani. Penelitian ini bertujuan untuk
mendeteksi dan mengetahui tingkat pembentukan biofilm pada enam isolat S.
aureus menggunakan Congo Red Agar dan Crystal Violet Binding Assay. Congo
Red Agar digunakan untuk melihat perubahan warna koloni, sedangkan Crystal
Violet Binding Assay digunakan untuk mengukur biomassa biofilm yang menempel
pada microplate berdasarkan nilai optical density (OD). Hasil pengujian dengan
Congo Red Agar menunjukkan bahwa isolat SA 3, SA 4, dan SA 5 positif
membentuk biofilm, sedangkan SA 6, SA 7, dan SA 8 menunjukkan hasil negatif.
Hasil pengujian dengan Crystal Violet Binding Assay menunjukkan bahwa seluruh
isolat membentuk biofilm. Isolat SA 3, SA 4, SA 7, dan SA 8 termasuk kategori
lemah, sedangkan SA 5 dan SA 6 termasuk kategori sedang berdasarkan
perbandingan nilai OD dengan nilai OD cut-off. Studi ini menunjukkan bahwa
semua isolat mampu membentuk biofilm dengan hasil sensitivitas yang berbeda
untuk kedua metode. Crystal Violet Binding Assay memiliki sensitivitas yang lebih
baik dibandingkan Congo Red Agar dalam mendeteksi biofilm. | |
| dc.description.abstract | Staphylococcus aureus is a bacterium capable of forming biofilms, which can
make infections more difficult to treat. This study aimed to detect and determine
the level of biofilm formation in six S. aureus isolates using Congo Red Agar and
the Crystal Violet Binding Assay. Congo Red Agar was used to observe changes in
colony color, while the Crystal Violet Binding Assay was used to measure the
biomass of biofilms attached to microplates based on optical density (OD) values.
The Congo Red Agar test showed that isolates SA 3, SA 4, and SA 5 were positive
for biofilm formation, whereas SA 6, SA 7, and SA 8 showed negative results. The
Crystal Violet Binding Assay showed that all isolates were capable of forming
biofilms. Isolates SA 3, SA 4, SA 7, and SA 8 were classified as weak biofilm
producers, while SA 5 and SA 6 were classified as moderate biofilm producers
based on comparisons between their OD values and the OD cut-off value. This
study demonstrated that all isolates were capable of forming biofilms, although the
two methods showed different levels of sensitivity. The Crystal Violet Binding
Assay was more sensitive than Congo Red Agar in detecting biofilm formation. | |
