dc.description.abstractKeadaan sirih merah (Piper crocatum) masih cukup terbatas dalam basis data global seperti NCBI. Penelitian ini bertujuan dalam menganalisis kekerabatan serta keragaman genetik antar individu sirih merah yang berbeda berdasarkan wilayahnya di beberapa daerah Indonesia, yaitu Bogor, Bandung, Sukabumi, Bekasi, dan Banten (Pandeglang) dengan analisis molekuler Sequence Related Amplified Polymorphism (SRAP). Metode penelitian meliputi isolasi DNA genom, kuantifikasi DNA, PCR-SRAP, dan analisis data. Hasil penelitian menunjukkan nilai normal pada konsentrasi dan kemurnian DNA. Optimasi 16 kombinasi primer diujikan dan terpilih primer kombinasi 1 (Me1-Em1) dengan suhu annealing terbaik di fase I sebesar 45 °C dan fase II sebesar 58 °C. Analisis data filogenetik dengan metode UPGMA dan koefisien Dice menunjukkan kemiripan genetik berkisar 0,82–1,00. Kesimpulannya, sirih merah asal Sukabumi dan Bekasi mempunyai nilai identik (1,00). Sebaliknya sirih merah asal Banten (Pandeglang) tergolong outgroup sebagai pemilik polimorfisme terbaik.
dc.description.abstractInformation regarding Red Betel (Piper crocatum) remains significantly limited in global databases such as NCBI. This study aims to analyze the genetic relationships and diversity among red betel individuals from various regions in Indonesia, specifically Bogor, Bandung, Sukabumi, Bekasi, and Banten(Pandeglang), using Sequence-Related Amplified Polymorphism (SRAP) molecular analysis. The research methods included genomic DNA isolation, DNA quantification, PCR-SRAP, and data analysis. The results indicated normal values for DNA concentration and purity. Optimization of 16 primer combinations was conducted, resulting in the selection of primer combination 1 (Me1-Em1), with an optimal annealing temperature of 45 °C for Phase I and 58 °C for Phase II. Phylogenetic data analysis using the UPGMA method and Dice coefficient showed genetic similarity ranging from 0.82 to 1.00. In conclusion, red betel accessions from Sukabumi and Bekasi exhibited identical values (1.00). Conversely, the red betel from Banten (Pandeglang) was categorized as an outgroup, demonstrating the highest level of polymorphism among the studied accession.
