      Karakterisasi Aktivitas Bakteri Denitrifikasi sebagai Agen Probiotik di Tambak Udang

      Abstract
      Akumulasi senyawa nitrogen toksik pada sistem budidaya udang intensif menjadi faktor pembatas utama kualitas air dan produktivitas. Pendekatan berbasis bakteri denitrifikasi berpotensi menjadi solusi biologis yang efisien, namun karakterisasi aktivitas dan kinerjanya masih terbilang terbatas. Penelitian ini bertujuan mengkarakterisasi aktivitas, kinetika, dan potensi bakteri denitrifikasi sebagai agen probiotik. Isolat ASLT, ASLT2, B25, dan B27 dianalisis melalui karakterisasi morfologi, kurva pertumbuhan, uji reduksi nitrogen, serta pendekatan kinetika Michaelis-Menten dan identifikasi molekuler berbasis gen 16S rRNA. Hasil ditunjukkan dengan isolat ASLT dan B27 yang memiliki kinerja denitrifikasi dominan dengan efisiensi reduksi nitrat >84% dan akumulasi nitrit sangat rendah, berkisar <1%. Analisis kinetika pada ASLT menunjukkan performa maksimal dengan afinitas substrat lebih tinggi (Km 0,52 mM) dan kapasitas reaksi maksimum lebih besar (Vmax 0,64 mM). Analisis filogenetik mengidentifikasi ASLT berkerabat dengan Staphylococcus sp. dan B27 dengan Bacillus altitudinis. Hasil ini menegaskan bahwa isolat ASLT unggul secara aktivitas dan efisiensi metabolik, sedangkan B27 prospektif untuk diaplikasikan. Studi ini memperkuat dasar pemanfaatan bakteri denitrifikasi sebagai strategi probiotik untuk pengelolaan nitrogen dan peningkatan kualitas tambak udang.
       
      The accumulation of toxic nitrogen compounds in intensive shrimp aquaculture systems is a major constraint on water quality and productivity. Denitrifying bacteria represent a promising biological solution. However, their functional performance and kinetic characteristics remain insufficiently explored. This study aimed to characterize the activity, kinetics, and probiotic potential of denitrifying bacteria. Isolates ASLT, ASLT2, B25, and B27 were evaluated through morphological characterization, growth profiling, nitrogen reduction assays, Michaelis-Menten kinetic analysis, and 16S rRNA-based molecular identification. The results showed that ASLT and B27 exhibited dominant denitrification performance, with nitrate reduction efficiencies exceeding 84% and deficient nitrite accumulation <1%. Kinetic analysis revealed that ASLT demonstrated superior performance, with higher substrate affinity (Km 0,52 mM) and greater maximum reaction rate (Vmax 0,64 mM). Phylogenetic analysis identified ASLT as closely related to Staphylococcus sp. and B27 to Bacillus altitudinis. These findings confirm that ASLT is superior in terms of activity and metabolic efficiency, while B27 shows greater potential for practical application. Overall, this study provides a strong foundation for the development of denitrifying bacteria as probiotic agents for nitrogen management and the improvement of shrimp pond water quality.
