The miana plant (Coleus scutellarioides (L.) Benth) contains flavonoid compounds that have potential as natural antioxidants. This study aimed to measure the flavonoid content and antioxidant capacity of miana leaves under shading levels of 0%, 25%, 50%, and 75%. Flavonoid content was analyzed using the aluminum chloride calorimetry (AlCl3) method, while antioxidant capacity was determined using the 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and cupric ion reducing antioxidant capacity (CUPRAC) methods. The results showed that 25% shading had the highest flavonoid content of 1.116 ± 0.012 mg QE/g DW, while the highest antioxidant capacity was obtained at 0% shading at 1.979 ± 0.006 µM TE/g DW (ABTS) and 184.427 ± 2.894 µM TE/g DW (CUPRAC). The higher antioxidant capacity values in the CUPRAC method indicate that this method is more representative in reflecting the antioxidant capacity of miana leaves. A positive correlation was observed between flavonoid content and both ABTS and CUPRAC antioxidant capacities under 25% and 50% shading treatments, while a negative correlation was observed under 0% and 75% shading treatments. This indicates that light intensity affects the content of bioactive compounds in miana leaves.
