      Abstract
      Timbunan sampah organik Rumah Kompos IPB University menyimpan gen-gen fungsional yang penting bagi pengembangan biokonversi lignoselulosa, namun belum pernah dikaji. Penelitian ini bertujuan mengidentifikasi struktur komunitas mikrobioma dan gen fungsional sampel kompos menggunakan teknik whole metagenome sequencing. Tahapan meliputi sampling dari empat titik, ekstraksi DNA, quality control, sekuensing, dan analisis bioinformatika. Terdapat pola suksesi komunitas dari filum Actinobacteria pada fase bahan mentah ke Proteobacteria dan Firmicutes pada fase pengomposan dan pematangan. Rekonstruksi dua genom MAG KMP.6 lebih unggul dalam potensi hidrolisis polisakarida melalui tingginya Glycoside Hydrolases dan Carbohydrate-Binding Modules, sedangkan MAG KMP.23 terspesialisasi pada degradasi polimer asam uronat melalui Polysaccharide Lyases serta biosintesis seluler melalui Glycosyl Transferases. Identifikasi spesies mengungkap kekerabatan KMP.23 dengan Ohtaekwangia koreensis DSM 25262 yang diisolasi dari pantai Korea Selatan, sedangkan KMP.6 dengan strain termofilik Paraflavitalea speifideaquila BL16E dari gua lava Hawaii.
       
      The organic waste at IPB University's Rumah Kompos harbors unstudied functional genes relevant to lignocellulose bioconversion. Whole metagenome sequencing of four compost samples, DNA extraction, quality control, sequencing, and bioinformatics analysis identified a microbial succession from Actinobacteria in the raw material phase to Proteobacteria and Firmicutes during maturation. Reconstruction of two MAGs revealed distinct functional profiles: KMP.6 demonstrated superior polysaccharide hydrolysis potential via Glycoside Hydrolases and Carbohydrate-Binding Modules, while KMP.23 specialized in uronate polymer degradation via Polysaccharide Lyases and cellular biosynthesis via Glycosyl Transferases. Species identification linked KMP.23 to Ohtaekwangia koreensis DSM 25262 isolated from a South Korean beach, and KMP.6 to the thermophilic strain Paraflavitalea speifideaquila BL16E from a Hawaiian lava cave.
