      Analisis Keragaman Mutan Bawang Merah (Allium cepa var. aggregatum) Toleran Salinitas

      Abstract
      Salinitas merupakan salah satu faktor pembatas dalam budidaya bawang merah karena dapat menurunkan pertumbuhan dan hasil tanaman. Penelitian ini bertujuan menganalisis potensi toleransi salinitas pada genotipe mutan bawang merah hasil iradiasi sinar gamma melalui evaluasi morfologi, molekuler, dan seleksi ideotipe. Penelitian dilaksanakan di Kebun Percobaan Tajur 2 dan Laboratorium Pusat Kajian Hortikultura Tropika IPB University pada Agustus–Desember 2025. Bahan genetik yang digunakan berupa genotipe mutan varietas Sanren dan Lokananta hasil perlakuan iradiasi gamma dan seleksi cekaman salinitas. Analisis molekuler dilakukan menggunakan 13 primer Start Codon Targeted (SCoT). Hasil penelitian menunjukkan adanya keragaman morfologi pada karakter bentuk umbi, warna umbi, jumlah daun, dan bobot umbi per rumpun. Hasil penelitian menunjukkan induksi mutasi meningkatkan persentase genotipe toleran terhadap salinitas, yaitu dari 74% menjadi 81% pada varietas Sanren dan dari 35% menjadi 44% pada varietas Lokananta, dengan peningkatan masing-masing sebesar 7% dan 26%. Analisis molekuler menghasilkan tingkat polimorfisme sebesar 74,29% pada varietas Lokananta dan 77,78% pada varietas Sanren yang menunjukkan tingginya keragaman genetik antar genotipe. Analisis klaster membentuk dua kelompok utama sesuai varietas asalnya. Hasil analisis Mix Variable Selection Index (MVSI) menunjukkan genotipe S3 dan L4 sebagai genotipe terbaik sehingga berpotensi dikembangkan sebagai genotipe unggul toleran salinitas.
       
      Salinity is one of the limiting factors in shallot cultivation because it can inhibit growth and reduce yields. This study aims to analyze the potential for salinity tolerance in mutant genotypes of shallot produced by gamma-ray irradiation through morphological and molecular evaluations, as well as ideotype selection. The research was conducted at the Tajur 2 Experimental Farm and the Center for Tropical Horticulture Research Laboratory at IPB University from August to December 2025. The genetic materials used consisted of mutant genotypes of the Sanren and Lokananta varieties resulting from gamma irradiation treatment and salinity stress selection. Molecular analysis was performed using 13 Start Codon Targeted (SCoT) primers. The results showed morphological variation in bulb shape, bulb color, number of leaves, and bulb weight per plant. The results of the study show that mutation induction increased the percentage of salinity tolerance genotypes from 74% to 81% in the Sanren variety and from 35% to 44% in the Lokananta variety representing increases of 7% and 26%, respectively. Molecular analysis yielded a polymorphism level of 74.29% in the Lokananta variety and 77.78% in the Sanren variety, indicating high genetic diversity among genotypes. Cluster analysis identified two main groups based on their original varieties. The results of the Mix Variable Selection Index (MVSI) analysis indicate that genotypes S3 and L4 are the best genotypes and thus have the potential to be developed as superior salinity tolerance genotypes.
