      Identifikasi, Karakterisasi, dan Analisis Potensi Dark Septate Endophyte dari Akar Digitaria ciliaris sebagai Agen Revegetasi Lahan Pascatambang Timah Belitung

      Abstract
      Degradasi lahan global mencapai sekitar 33% dari total daratan, dengan Indonesia menyumbang sekitar 48,3 juta hektar lahan terdegradasi. Kabupaten Belitung Timur mengalami peningkatan kerusakan hutan dari 33.286 hektar pada tahun 2013 menjadi 61.033 hektar pada tahun 2018 akibat aktivitas pertambangan timah. Aktivitas ini menghilangkan vegetasi dan menyisakan tailing berpasir yang menyebabkan rendahnya kapasitas tanah dalam menyimpan air dan hara. Sebagai upaya revegetasi, Fungi Dark Septate Endophyte (DSE) berpotensi dimanfaatkan sebagai agen hayati yang mampu meningkatkan toleransi tanaman terhadap cekaman lingkungan dan serapan hara. Penelitian ini bertujuan mengkarakterisasi dan menganalisis potensi isolat DSE yang diisolasi dari akar Digitaria ciliaris untuk mendukung revegetasi lahan pascatambang timah di Belitung. Sebanyak 12 isolat DSE berhasil diisolasi dari akar, daun, dan batang semu D. ciliaris. Berdasarkan hasil kolonisasi yang dominan pada akar, empat isolat asal akar dipilih untuk karakterisasi morfologi serta pengujian kemampuan pelarutan fosfat, aktivitas hemolisis, dan patogenisitas pada benih jagung dan trembesi. Seluruh isolat akar teridentifikasi sebagai genus Curvularia. Isolat Curvularia sp. A3 menunjukkan indeks pelarutan fosfat tertinggi (0,75), tidak bersifat hemolitik, serta menghasilkan tinggi tanaman tertinggi pada jagung (7,76 cm) dan trembesi (10,25 cm). Hasil penelitian menunjukkan bahwa Curvularia sp. A3 berpotensi dikembangkan sebagai agen hayati untuk mendukung revegetasi lahan pascatambang timah.
       
      Global land degradation affects approximately 33% of the world's terrestrial area, with Indonesia contributing around 48.3 million hectares of degraded land. In East Belitung Regency, forest degradation increased from 33,286 ha in 2013 to 61,033 ha in 2018 due to tin mining activities. Mining removes natural vegetation and leaves sandy tailings with low water and nutrient retention capacity. Dark Septate Endophytic fungi (DSE) have potential as biological agents to enhance plant tolerance to environmental stress and improve nutrient uptake. This study aimed to characterize and evaluate the potential of DSE isolates obtained from the roots of Digitaria ciliaris for revegetation of post-tin mining land in Belitung. A total of 12 DSE isolates were obtained from roots, leaves, and pseudostems of D. ciliaris. Based on the higher colonization observed in roots, four root-derived isolates were selected for morphological characterization and assessment of phosphate-solubilizing ability, hemolytic activity, and pathogenicity on maize and rain tree seedlings. All root isolates were identified as Curvularia. Curvularia sp. A3 exhibited the highest phosphate-solubilization index (0.75), showed no hemolytic activity, and produced the greatest plant height in maize (7.76 cm) and rain tree seedlings (10.25 cm). These findings indicate that Curvularia sp. A3 is a promising biological agent for revegetation of post-tin mining land.
