      Aplikasi Berbagai Jenis Media Dasar dan Konsentrasi BAP terhadap Induksi Protocorm Like Bodies Cattleya Hibrida var. Sophia Martin secara In Vitro

      Abstract
      Tingginya keragaman spesies anggrek di Indonesia menjadikan tanaman hias ini diminati oleh penggemar tanaman hias dalam negeri maupun luar negeri, salah satunya Cattleya Sophia Martin. Namun pembibitan secara konvensional kurang optimal sehingga memerlukan waktu lama dan hasilnya tidak seragam. Oleh karena itu, perbanyakan secara in vitro merupakan solusi atas permasalahan tersebut. Penelitian ini bertujuan memperlajari pengaruh jenis media dasar dan konsentrasi penggunaan 6-benzylaminopurine (BAP) terhadap induksi protocorm like bodies (PLB) secara in vitro untuk menghasilkan perbanyakan bibit anggrek yang optimum. Percobaan disusun dalam Rancangan Kelompok Lengkap Teracak (RKLT) dengan 2 faktor perlakuan, yang terdiri dari 24 kelompok perlakuan, dan 4 ulangan, sehingga terdapat 96 percobaan. Faktor pertama adalah jenis media dasar yang digunakan, yaitu Knudson C (KC), Schenk and Hildebrant (SH), dan Vacin and Went (VW) dengan dua taraf konsentrasi hara makro dan hara mikro, yaitu ½ dan ¾ konsentrasi. Faktor kedua yaitu penambahan BAP dengan empat taraf konsentrasi (0; 1; 2; 3 mg L-1 ). Perlakuan tunggal jenis media SH ¾ menghasilkan rata-rata PLB per eksplan tertinggi yang berbeda nyata pada 20 minggu setelah kultur (MSK) yaitu sebanyak 4,04 PLB per eksplan. Perlakuan tunggal penambahan BAP 2 mg L-1 menghasilkan rata-rata PLB per eksplan tertinggi yang berbeda nyata pada 12 MSK yaitu sebanyak 2,71 PLB per eksplan. Pengaruh tunggal konsentrasi BAP memberikan hasil yang berpengaruh nyata lebih cepat dibandingkan dengan perlakuan tunggal jenis media. Hasil penelitian ini memperoleh kombinasi perlakuan pada media kultur yang dapat dijadikan rekomendasi media pembentukan tiap parameter yang telah diamati.
       
      Indonesia’s high orchid diversity has attracted both domestic and international ornamental plant enthusiasts, including Cattleya Sophia Martin. However, conventional propagation methods are inefficient due to long production times and inconsistent seedling quality. Therefore, in vitro propagation offers a more effective alternative. This study aimed to examine the effects of basal media types and concentration of 6-benzylaminopurine (BAP) on the in vitro induction of protocorm like bodies (PLBs). The experiment was conducted using a Randomized Complete Block Design with two factors, 24 treatment combinations, and four replications. Basal media consisted of Knudson C (KC), Schenk and Hilderbrandt (SH), and Vacint and Went (VW), each applied at ½ and ¾ nutrient strength. BAP was added at four concentration (0; 1; 2; 3 mg L-1 ). The highest PLB production was obtained on SH medium at ¾ strength nutrition, yielding an average of 4,04 PLBs per explant at 20 weeks after culture. BAP at 2 mg L-1 produced the highest PLB number in earliest observation time, with an average of 2.71 PLBs per explant at 12 weeks after culture. The single effect of BAP concentration influenced PLB induction earlier than the basal media effect and the interaction between the two treatments was not statistically significant. This study obtained a combination of treatments in culture media that can be recommended for the induction of each observed growth parameter.
