      Perubahan Senyawa Bioaktif Teh Tubruk Komersial: Investigasi Berdasarkan Faktor Suhu dan Durasi Penyeduhan

      Abstract
      Teh tubruk merupakan salah satu bentuk teh konsumsi yang populer di Indonesia, namun informasi mengenai metode penyeduhan yang optimal untuk mempertahankan kandungan senyawa bioaktifnya masih terbatas. Penelitian ini bertujuan menentukan pengaruh variasi suhu (62 oC, 77 oC, dan 85 oC) dan durasi penyeduhan (15, 120, dan 300 detik) terhadap kandungan senyawa bioaktif dan kapasitas antioksidan seduhan teh hijau melati tubruk merek TongTji. Parameter yang dianalisis meliputi kadar total fenolik, kadar asam galat, katekin, dan kafein, serta kapasitas antioksidan metode DPPH dan FRAP. Data dianalisis menggunakan two-way ANOVA dan uji lanjut Tukey’s HSD (p < 0,05). Hasil menunjukkan bahwa suhu dan durasi penyeduhan berpengaruh signifikan terhadap seluruh parameter yang diuji. Kadar fenolik total tertinggi diperoleh pada suhu 77 oC selama 300 detik (0,8916 mg GAE/mL), sedangkan kapasitas antioksidan tertinggi diperoleh pada suhu 85 oC selama 300 detik (2,0030 mg AAE/mL metode DPPH dan 2,3361 mg AAE/mL metode FRAP) yang tidak berbeda nyata dengan suhu 77 oC pada durasi yang sama. Analisis korelasi menunjukkan bahwa kapasitas antioksidan tidak hanya dipengaruhi oleh total fenolik, namun juga komponen bioaktif spesifik. Secara keseluruhan, peningkatan suhu dan durasi penyeduhan meningkatkan ekstraksi senyawa bioaktif dengan kondisi optimal pada 77–85 oC durasi 300 detik.
       
      Tubruk tea or loose-leaf tea is one of the most consumed tea forms in Indonesia, however information on optimal brewing to preserve its bioactive compounds remains limited. This study aimed to determine the effects of brewing temperature (62 oC, 77 oC, and 85 oC) and duration (15, 120, and 300 seconds) on the bioactive compound content and antioxidant capacity of TongTji green jasmine loose-leaf tea infusion. Parameters included total phenolic content, gallic acid, catechin, and caffeine levels, and antioxidant capacity (DPPH and FRAP). Data were analyzed using two-way ANOVA followed by Tukey’s HSD test (p < 0.05). Results showed that brewing temperature and duration significantly affected all parameters. The highest total phenolic content was obtained at 77 oC for 300 seconds (0.8916 mg GAE/mL), while the highest antioxidant capacity was achieved at 85 oC for 300 seconds (DPPH: 2.0030 mg AAE/mL and FRAP: 2.3361 mg AAE/mL), not significantly different from 77 oC at the same duration. Correlation analysis indicated that antioxidant capacity was influenced not only by total phenolic content but also by specific bioactive compounds. Overall, higher temperature and longer duration enhanced bioactive compound extraction, with optimal brewing conditions at 77 – 85 oC for 300 seconds.
