      Pengaruh Jenis dan Konsentrasi Auksin terhadap Induksi Perakaran Lavender (Lavandula angustifolia) Secara In Vitro

      Abstract
      Lavender (Lavandula angustifolia) merupakan tanaman penghasil minyak atsiri yang digunakan sebagai bahan baku kosmetik, obat-obatan, dan parfum. Tingginya permintaan pasar perlu didukung dengan penyediaan bibit salah satunya melalui perbanyakan lavender secara kultur in vitro. Salah satu tahapan penting dalam perbanyakan in vitro lavender adalah pengakaran tunas. Penelitian ini bertujuan mengetahui pengaruh pemberian konsentrasi IBA dan NAA pada laju pertumbuhan dan perkembangan akar pada tunas lavender. Penelitian ini dilakukan pada Oktober 2024 hingga Maret 2025 di Laboratorium Biologi Molekuler Tanaman 1, Departemen Agronomi dan Hortikultura, Fakultas Pertanian IPB dengan menggunakan Rancangan Kelompok Lengkap Teracak dua faktor. Bahan yang digunakan untuk percobaan induksi akar adalah tunas Lavandula angustifolia dengan media tanpa auksin sebagai kontrol, 0,3 ppm NAA, 0,4 ppm IBA, 0,6 ppm IBA, 0,3 ppm NAA + 0,4 ppm IBA, dan 0,3 ppm NAA + 0,6 ppm IBA. Pengamatan dilakukan terhadap karakter morfologi jumlah akar, tunas, daun, buku, jumlah tunas membentuk kalus, dan persentase tunas berkalus. Hasil dari percobaan menunjukkan pemberian kombinasi auksin pada tunas lavender tidak memberikan hasil yang nyata terhadap jumlah akar, namun meningkatkan karakter jumlah tunas membentuk kalus. Kombinasi media 0,3 ppm NAA + 0,6 ppm IBA menghasilkan pertumbuhan jumlah tunas membentuk kalus terbanyak. Perlakuan 0,4 ppm IBA merupakan media terbaik untuk karakter jumlah tunas, buku, dan daun.
       
      Lavender (Lavandula angustifolia) is an essential oil-producing plant widely used as a raw material in cosmetics, pharmaceuticals, and perfumes. The high market demand for lavender necessitates a sufficient supply of seedlings, which can be achieved through in vitro propagation. One of the critical stages in lavender micropropagation is root induction. This study aimed to investigate the effects of different concentrations of Indole-3-Butyric Acid (IBA) and 1-Naphthaleneacetic Acid (NAA) on root growth and development in lavender shoots. The research was conducted from October 2024 to March 2025 at the Plant Molecular Biology Laboratory 1, Department of Agronomy and Horticulture, Faculty of Agriculture, IPB University, using a two factor Randomized Complete Block Design (RCBD). The plant material used in the root induction experiment consisted of Lavandula angustifolia shoots. Treatments included an auxin-free control, 0,3 ppm, 0,4 ppm IBA, 0,6 ppm IBA, and combinations of 0,3 ppm NAA + 0,4 ppm IBA, and 0,3 ppm NAA + 0,6 ppm IBA. Morphological observations were made on root number, shoot number, leaf number, number of nodes, number of shoots forming callus, and percentage of callused shoots. The results showed that auxin combinations had no significant effect on root number but increased the number of shoots forming callus. The combination of 0,3 ppm NAA + 0,6 ppm IBA produced the highest number of callus forming shoots. The 0,4 ppm IBA treatment was found to be optimal for increasing shoot number, node number, and leaf number.
