      Pemurnian dan Karakterisasi Bakteriosin dari Nata de Coco dan Limbah Cair Fermentasi

      Abstract
      Komagataeibacter xylinus, bakteri utama dalam fermentasi nata de coco, diprediksi oleh perangkat lunak antiSMASH (antibiotics and secondary metabolite analysis shell) mampu menghasilkan bakteriosin. Bakteriosin telah dilaporkan berpotensi sebagai kandidat antibiotik alami untuk mengatasi resistensi antibiotik. Penelitian ini bertujuan memurnikan dan mengkarakterisasi bakteriosin dari K. xylinus untuk membuktikan prediksi in silico oleh antiSMASH secara in vitro. Bakteriosin dari nata de coco dan limbah cair fermentasi dimurnikan secara parsial melalui fraksinasi amonium sulfat bertingkat dan kadar proteinnya diukur dengan metode Bradford. Berat molekul bakteriosin dianalisis menggunakan SDS-PAGE dan aktivitas antibakterinya diuji terhadap Escherichia coli ATCC 8739 dan Staphylococcus aureus ATCC 6538. Fraksi amonium sulfat 60% dari nata de coco dan limbah cair fermentasi menghasilkan pita protein tunggal dengan berat molekul sekitar 15 kDa dan memiliki aktivitas antibakteri bersifat bakteriostatik terhadap S. aureus ATCC 6538, tetapi tidak menunjukkan aktivitas antibakteri terhadap E. coli ATCC 8739. Hasil penelitian ini mendukung prediksi in silico oleh antiSMASH.
       
      Komagataeibacter xylinus, the primary bacterium involved in nata de coco fermentation, is predicted by the antiSMASH (antibiotics and secondary metabolite analysis shell) software to produce bacteriocins. Bacteriocins have been reported as potential natural antibiotic candidates for combat antibiotic resistance. This study aimed to purify and characterize bacteriocins from K. xylinus to verify the antiSMASH in silico prediction through in vitro analysis. Bacteriocins from nata de coco and fermentation liquid waste were partially purified using stepwise ammonium sulfate fractionation, and the protein concentration was measured using the Bradford method. The molecular weight of the bacteriocins was analyzed using SDS-PAGE, and antibacterial activity was tested against Escherichia coli ATCC 8739 and Staphylococcus aureus ATCC 6538. The 60% ammonium sulfate fraction from both nata de coco and fermentation liquid waste exhibited a single protein band with an approximate molecular weight of 15 kDa and showed bacteriostatic antibacterial activity against S. aureus ATCC 6538, but no activity against E. coli ATCC 8739. These results support the in silico prediction by antiSMASH.
