      Uji Cepat Viabilitas Benih Umbi Bawang Putih (Allium sativum L.) dengan Tetrazolium

      Abstract
      Konsumsi bawang putih yang semakin meningkat setiap tahun perlu diikuti dengan mutu benih yang digunakan dalam menunjang produksi bawang putih. Indikator pemeriksaan mutu benih bawang putih yang cepat dan efektif diharapkan dapat menjamin viabilitas benih. Penelitian ini bertujuan menentukan metode konsentrasi dan lama inkubasi terhadap standar evaluasi pola pewarnaan pengujian TTZ. Benih yang digunakan yaitu benih umbi bawang putih varietas Lumbu Putih dan dibuat menjadi 3 lot yaitu benih segar (viabilitas tinggi), benih yang diusangkan suhu 45 oC 48 jam (viabilitas sedang) dan suhu 50 oC 48 jam (viabilitas rendah). Rancangan yang digunakan untuk pengujian tetrazolium yaitu RKLT 2 faktor. Faktor yang diamati adalah 2 taraf konsentrasi dan 2 taraf lama inkubasi. Konsentrasi yang digunakan adalah 0,25%, 0,5% yang diinkubasi selama 1 jam dan 2 jam. Rancangan yang digunakan dalam uji mutu fisiologis yaitu RKLT 1 faktor. Faktor yang diamati yaitu lot benih umbi bawang putih. Metode terbaik untuk uji TTZ adalah dengan menggunakan konsentrasi 0,25% dan inkubasi 1 jam. Evaluasi pewarnaan untuk deteksi viabilitas menghasilkan 10 pola pewarnaan benih viabel dan 7 pola pewarnaan benih non-viabel. Hasil pengujian menggunakan tetrazolium ini efektif dalam menilai viabilitas benih umbi bawang putih serta memiliki korelasi erat dengan pengujian mutu fisiologis berdasar daya tumbuh, tinggi tanaman, benih segar, kecepatan tumbuh, dan potensi tumbuh maksimum.
       
      The increasing consumption of garlic every year needs to be followed by the quality of the seeds used to support garlic production. A quick and effective seed quality testing is expected to ensure the viability of the seeds. This study aims to determine the concentration method and incubation time for the standard evaluation of staining patterns for the TTZ testing. The seeds used were garlic bulb seeds of the Lumbu Putih variety and were divided into 3 lots: fresh seeds (high viablitity), seeds exposed to 45 °C for 48 hours (medium viability), and seeds exposed to 50 °C for 48 hours(low viability). The design used for the physiological test was a Completely Randomized Block Design (RKLT) with two factors. The tetrazolium test was based on two concentration levels and two incubation duration levels. The concentrations used were 0.25% and 0.5%, with incubation times of 1 hour and 2 hours. The design used for the physiological test was a Completely Randomized Block Design (RKLT) with one factor. The factor observed was the garlic bulb seed lot. The result showed that the best method for the tetrazolium test is using a concentration of 0.25% and incubating for 1 hour. The staining evaluation for viability detection showed 10 viable seed staining patterns and 7 non-viable seed staining patterns. The tetrazolium test results were effective in assessing the viability of garlic bulb seeds and showed a strong correlation with physiological quality testing in the form of growth power, plant height, fresh seedlings, growth speed, and maximum growth potential.
