      Induksi Kalus dan Regenerasi Tanaman Temu Putih (Curcuma zedoaria) menggunakan 2,4-D dan BAP dengan Metode Thin Cell Layer (TCL)

      Abstract
      Temu putih (Curcuma zedoaria) merupakan tanaman biofarmaka yang dimanfaatkan sebagai rempah dan obat herbal. Induksi kalus dan regenerasi menggunakan potongan Thin Cell Layer (TCL) dari pangkal batang mampu menghasilkan tanaman berkualitas tinggi dalam jumlah besar dengan waktu yang lebih singkat. Penelitian ini bertujuan mempelajari pengaruh pemberian 2,4-D dan BAP terhadap pembentukan kalus dan regenerasi pada tanaman temu putih menggunakan metode TCL secara in vitro. Penelitian dilakukan pada bulan Maret hingga Desember 2023 di Laboratorium Plant Molecular Biology 1, Departemen Agronomi dan Hortikultura, Fakultas Pertanian, IPB University. Penelitian ini menggunakan RKLT faktorial dengan dua faktor. Faktor pertama adalah 2,4-D (0, 1, 2, dan 3 mg L-1) dan faktor kedua adalah BAP (0, 0,5, 1 mg L-1). Hasil penelitian menunjukkan pemberian 2,4-D 0 mg L-1 dan BAP 1 mg L-1 secara tunggal memberikan hasil paling optimal untuk menginduksi kalus. Warna kalus temu putih diamati terbanyak dimiliki kode 2.5Y 7/2 (putih krem keabuan) (19,2%) dan dominan berstruktur remah (57,8%). Interaksi 2,4-D dan BAP memberikan pengaruh yang signifikan terhadap proses regenerasi. Pemberian BAP 0,5 mg L-1 tanpa penambahan 2,4-D optimal menginisiasi regenerasi eksplan per botol serta meningkatkan jumlah akar, planlet, dan daun. Pemberian 2,4-D 0 mg L-1 dan BAP 1 mg L-1 optimal menginisiasi waktu muncul tunas dan meningkatkan jumlah tunas.
       
      White turmeric (Curcuma zedoaria) is a biopharmaceutical plant used as a spice and herbal medicine. Callus induction and regeneration using Thin Cell Layer (TCL) explants from the stem bases produce high-quality and high-quantity plantlets in a short time. This study aimed to investigate the effect of 2,4-D and BAP on callus formation and shoot regeneration of white turmeric plants using an in vitro TCL method. This study was conducted from March to December 2023 at the Plant Molecular Biology 1 Laboratory, Department of Agronomy and Horticulture, Faculty of Agriculture, IPB University. This study used an RKLT factorial with two factors. The first factor was 2,4-D (0, 1, 2, and 3 mg L-1) and the second factor was BAP (0, 0.5, and 1 mg L-1). The results showed that a single application of BAP 1 mg L-1 without 2,4-D was the best for inducing callus formation. Most of the observed white turmeric callus colors was color code 2.5Y 7/2 (grayish creamy white) (19.2%) and had crumb structures (57.8%). The interaction between 2,4-D and BAP significantly affects on the regeneration process. Application of BAP 0.5 mg L-1 without 2,4-D optimally initiated explant regeneration per bottle and increased the root, planlets, and leaf numbers. Meanwhile, 2,4-D 0 mg L-1 and BAP 1 mg L-1 optimally initiated shoot emergence and increased the number of shoots.
