The Construction of pCAMBIA 1303-Stilbene Syntase Recombinant DNA Preventive for Rotten Root Disease of Oil Palm
Abstract
An oil palm is one of the important estate commodities in Indonesia. Nevertheless the effort to increase its productivity has many hindrances. One of them is rotten root disease caused by Ganoderma spp. New approach to get resistence oil palm toward Ganoderma attack is to insert gene produces substance played role as an antifungi. One of genes that produces antifungi is stilbene syntase gene derived from grapes (Vitis vinifera). The purpose of this research is to cloning stilbene syntase gene on to pCAMBIA 1303 expression vector between 35S CaMV promoter. Stilbene syntase gene was cloned on to pCAMBIA 1303 expression vector then to be transformed on to Escherichia coli XL-1 Blue and the last to be inserted on to Agrobacterium tumefaciens. The result of this research shows stilbene syntase gene was successful to be cloned to pCAMBIA 1303. This result was showed by two band sized 12000 bp (pCAMBIA 1303 plasmid) and 1500 bp (stilbene syntase gene) in agarose gel after have been restricted with SpeI and NcoI enzyme.
