      Induksi Hemoglobin Fetal pada Sel K562 Berbasis Herbal Seledri (Apium graveolens) dan Alang-Alang (Imperata cylindrica) untuk Talasemia Beta

      Abstract
      Talasemia-β merupakan penyakit genetik yang disebabkan oleh tidak normalnya sintesis rantai globin-β, sehingga mengakibatkan ketidakseimbangan rantai globin. Salah satu pendekatan terapi yang aman adalah dengan stimulasi sintesis rantai globin-γ untuk mengompensasi ketidakseimbangan rantai globin dengan membentuk Hemoglobin Fetal (HbF). Penelitian ini bertujuan menganalisis pengaruh perlakuan masing-masing ekstrak herbal A. graveolens (AG) dan I. cylindrica (IC) terhadap viabilitas sel K562, produksi Hb total, dan induksi HbF. Metode untuk menentukan sel yang mengandung hemoglobin dilakukan melalui pewarnaan dengan uji benzidin, dilanjutkan dengan pengukuran sel yang terinduksi HbF dengan metode ELISA. Hasil penelitian menunjukkan bahwa ekstrak IC menghasilkan kadar konsentrasi HbF dengan nilai 2,691 ng/mL, lebih tinggi dibandingkan ekstrak AG dengan nilai 2,308 ng/mL. Kedua hasil perlakuan ekstrak tersebut masih berada di bawah kontrol positif hidroksiurea. Hasil ini mengindikasikan bahwa ekstrak IC lebih berpotensi dalam meringankan gejala klinis talasemia-β dibandingkan dengan ekstrak AG, meskipun potensi kedua ekstrak masih di bawah kontrol positif hidroksiurea.
       
      Thalassemia-β is a genetic disease caused by abnormal synthesis of the globin-β chain, resulting in an imbalance of globin chains. One safe therapeutic approach is to stimulate the synthesis of the globin-γ chain to counterbalance the globin chain imbalance by forming Fetal Hemoglobin (HbF). This study aimed to analyze the effects of treatment with each herbal extract of A. graveolens (AG) and I. cylindrica (IC) on the viability of K562 cells, total Hb production, and HbF induction. The method for determining cells containing hemoglobin was carried out through staining using the benzidine test, followed by measuring the HbF-induced cells using the ELISA method. Research results show that IC extract resulted in a concentration of HbF with a value of 2,691 ng/mL, higher than the value of 2,308 ng/mL obtained from AG extract. Both results of the extract treatments remained below the positive control of hydroxyurea. These findings suggest that IC extract holds more potential in alleviating clinical symptoms of thalassemia-β compared to AG extract, although the potential of both extracts still falls below the positive control of hydroxyurea.
