dc.description.abstractObesitas merupakan penyakit penyumbang kematian cukup tinggi di dunia dan angka penderitanya terus meningkat. Kemangi diketahui mengandung senyawa bioaktif yang baik bagi kesehatan namun potensinya sebagai antiobesitas melalui penghambatan enzim lipase pankreas belum diketahui. Penelitian bertujuan menguji potensi senyawa aktif daun kemangi sebagai inhibitor enzim lipase pankreas secara in silico. Metode LC-MS QTOF digunakan untuk identifikasi senyawa aktif, kemudian dilanjutkan dengan penapisan virtual, dan analisis bioavailabilitas dan toksisitas. Hasil penelitian menunjukkan 27 ligan uji hasil studi literatur dan 2 ligan hasil analisis LC-MS diketahui memiliki kemampuan yang baik dalam menghambat lipase pankreas (Kode: 1LPB) dibandingkan dengan orlistat berdasarkan energi bebas ikatan (ΔG), konstanta disosiasi (Kd), serta interaksi ligan-reseptor. Ligan uji alpha-amyrin merupakan ligan literatur yang memiliki penghambatan paling kuat dengan nilai ΔG -9,4800 kkal/mol dan konstanta disosiasi (Kd) 0,11 μM, serta lolos analisis bioavailabilitas dan toksisitas. Simpulan penelitian ini adalah daun kemangi diprediksi memiliki potensi sebagai antiobesitas melalui penghambatan lipase pankreas.id
dc.description.abstractObesity is a disease that is a fairly high contributor to death in the world and the number of sufferers continues to increase. Basil is known to contain bioactive compounds that are good for health, but their potential as an anti-obesity through inhibition of the pancreatic lipase enzyme is not yet known. This research aimed to test the potency of the active compound in basil leaves as an inhibitor of the pancreatic lipase enzyme in silico. The LC-MS QTOF method was used to identify the active compound, followed by virtual screening, and bioavailability and toxicity analysis. The results showed that 27 ligands as the result of the literature study and 2 ligands from LC-MS analysis were discovered to have good ability in inhibiting pancreatic lipase (Code: 1LPB) compared with orlistat based on the free energy of binding (ΔG), bioavailability and toxicity analysis, and ligand-receptor interactions. Ligand of alpha-amyrin had the strongest inhibition with the value ΔG -9,4800 kcal/mol and Kd of 0,11 μM, and passed the bioavailability and toxicity analysis. This study concludes that basil leaves were predicted to have potential as an anti-obesity agent through inhibition of pancreatic lipase.id
