      Identifikasi Senyawa Aktif Daun Kumis Kucing (Orthosiphon aristatus) dengan LC-MS dan Potensinya sebagai Inhibitor Lipase Pankreas In Silico

      Abstract
      Obesitas merupakan kondisi tubuh dengan penumpukan lemak berlebih yang menyebabkan tubuh mengalami kelebihan berat badan. Prevalensi obesitas di Indonesia dan dunia terus meningkat setiap tahunnya. Kumis kucing diketahui sebagai tanaman herbal yang berpotensi sebagai inhibitor lipase pankreas. Penelitian ini bertujuan mengidentifikasi senyawa aktif daun kumis kucing dengan LC-MS dan potensinya sebagai inhibitor lipase pankreas. Metode LC-MS digunakan untuk mengidentifikasi senyawa aktif dilanjutkan dengan penambatan molekuler menggunakan perangkat lunak YASARA. Hasil penelitian diperoleh 26 senyawa aktif dari ekstrak etanol 70% dan 12 ligan hasil studi literatur. Terdapat 14 senyawa yang berpotensi menjadi kandidat inhibitor lipase pankreas. Tiga senyawa menjadi kandidat terbaik inhibitor lipase pankreas, yaitu senyawa licoagroside A, tetramethylsutellarein serta satu senyawa penciri kumis kucing yaitu sinensetin, berdasarkan seluruh parameter uji.
       
      Obesity is a condition of the body with an excess of fat accumulation causing overweight. The prevalence of obesity in Indonesia and all over the world increases year by year. Cat’s whisker is known as herbal plant that has potential as pancreatic lipase inhibitor. This research aims to identify the active compounds of cat’s whisker leaves using LC-MS and its potential as a pancreatic lipase inhibitor. LC-MS method is used to identify the active compounds and followed by molecular docking with YASARA software. The result showed that there are 26 bioactive compounds from 70% ethanol extract and 12 bioactive compounds from literature study. There are 14 compounds, it’s can potentially become pancreatic lipase inhibitors. Licoagroside A, tetramethylscutellarein and sinensetin as one of the marker compound of cat’s whisker leaves are the most potential compounds as a pancreatic lipase inhibitor based on all parameters.
