      Karakteristik Basa Nukleotida Gen COI Rajungan Portunus pelagicus (Linnaeus, 1758) sebagai Dasar Pengelolaannya di WPP-NRI 711

      Abstract
      Sumberdaya perikanan Indonesia yang bersifat multispesies dapat menimbulkan fenomena cryptic dan complex species, sehingga dibutuhkan metode yang akurat dalam pemastian spesies rajungan, yaitu dengan DNA barcoding. Penelitian ini bertujuan menentukan kepastian spesies dan menganalisis kondisi populasi rajungan (Portunus pelagicus) di perairan WPP-NRI 711 berdasarkan gen COI. DNA contoh rajungan berjumlah 10 individu yang didapatkan dari tiga lokasi, yaitu perairan Bintan, Bangka, dan Ketapang telah diisolasi. Kualitas untaian DNA yang baik digandakan dan diurutkan pada proses sekuensing. Seluruh contoh rajungan tervalidasi secara genetik sebagai spesies Portunus pelagicus. Komposisi basa nukleotida dari contoh rajungan didominasi oleh pasangan basa adenin-timin. Keragaman genetik rajungan di perairan Ketapang dan Bintan tergolong tinggi, sedangkan di perairan Bangka tergolong sedang. Ketiga lokasi penelitian memiliki nilai Fst yang tergolong rendah. Hasil penelitian menunjukkan bahwa populasi rajungan berada pada kondisi yang baik.
       
      Indonesia's multi-species fisheries resources can cause cryptic and complex species phenomena, so an accurate method is needed to determine the blue swimming crab species, such as DNA barcoding. This study aimed to determine the species certainty and to analyze the population status of blue swimming crab (Portunus pelagicus) based on the COI gene in WPP-NRI 711. A total of 10 blue swimming crab samples DNA obtained from three locations such as Bintan, Bangka, and Ketapang waters was isolated. High quality DNA strands are duplicated and sequenced. All of the samples have been genetically validated as Portunus pelagicus species. The nucleotide base composition of the COI gene from samples was dominated by adenine-thymine pairs. The genetic diversity value in Ketapang and Bintan are high, while in Bangka is moderate. All sampling sites have a low fixation index value. It can be concluded that the blue swimming crab populations are in a good condition.
