      Autentikasi Teri Nasi dari Perairan Selat Madura berdasarkan Biomarka Gen COI Sebagai Dasar Pengelolaan

      Abstract
      Ikan teri nasi merupakan salah satu komoditas ekspor Indonesia. Ukuran Ikan yang kecil menyebabkan identifikasi morfologi kurang akurat sehingga diperlukan identifikasi secara biomolekuler untuk kepastian spesies. Penelitian ini bertujuan menentukan spesies ikan teri nasi dari perairan Selat Madura yakni di Pasuruan dan Sampang, Jawa Timur dengan biomarka Gen COI. Penelitian dilakukan pada bulan Januari hingga November 2020. Pengambilan contoh ikan teri dilakukan dengan bantuan nelayan menggunakan alat tangkap payang teri. Analisis molekuler meliputi ekstraksi DNA, amplifikasi DNA dan sekuensing. Berdasarkan hasil identifikasi secara morfologis, ikan teri nasi di perairan Lekok dan Camplong merupakan anggota dari Famili Engraulidae. Adapun secara molekuler berdasarkan gen COI, ikan teri nasi telah teridentifikasi sebagai Encrasicholina heteroloba dan E. pseudoheteroloba. Jarak genetik ikan teri E. pseudoheteroloba antara perairan Lekok dan Camplong adalah 0,007, sedangkan untuk spesies E. heteroloba adalah 0. Nilai jarak genetik ini menunjukkan hubungan kekerabatan yang sangat dekat dari masing-masing spesies teri nasi tersebut di perairan Lekok dan Camplong.
       
      Anchovy is one of Indonesia's export commodities. The small size of the fish causes inaccurate morphological identification, so biomolecular identification is required for species certainty. This study aims to determine the species of anchovies from the waters of the Madura Strait, namely in Pasuruan and Sampang, East Java, using COI gene biomarkers. The research was conducted from January to November 2020. A sampling of anchovies was carried out with the help of fishermen using payang anchovies fishing gear. The molecular analysis includes DNA extraction, DNA amplification, and sequencing. Based on the results of morphological identification, anchovy in Lekok and Camplong waters is a members of the Engraulidae family. Molecularly based on the COI gene, anchovy rice has been identified as Encrasicholina heteroloba and E. pseudoheteroloba. The genetic distance for anchovy E. pseudoheteroloba between Lekok and Camplong waters was 0.007, while for E. heteroloba species it was 0. The value of this genetic distance indicated a very close kinship of each anchovy species in Lekok and Camplong waters.
