Complete blood count (CBC) merupakan salah satu pemeriksaan darah yang
dilakukan untuk mengevaluasi kesehatan ternak. Beberapa faktor pra-analitik yang
turut berpengaruh terhadap data yang dihasilkan di antaranya jenis antikoagulan
yang digunakan, jeda waktu pemeriksaan, serta suhu penyimpanan. Oleh karena itu
faktor tersebut penting dan perlu diperhatikan pada saat sebelum pemeriksaan darah
dilakukan. Penelitian ini bertujuan untuk mengetahui jumlah dan morfologi leukosit
darah yang diberi antikoagulan Tripotassium Ethylene Diamine Tetraacetic Acid
(K3EDTA) dan disimpan selama 48 jam pada suhu 4 °C. Hewan yang digunakan
adalah 10 ekor sapi peranakan ongole (PO) betina dengan umur 4 - 5 tahun. Sampel
darah diambil melalui vena jugularis untuk diamati jumlah dan morfologi jenis sel
leukosit dengan waktu penyimpanan 0, 3, 6, 9, 12, 24, dan 48 jam. Hasil penelitian
menunjukkan jumlah total leukosit, limfosit, neutrofil, eosinofil, dan basofil sampai
pengamatan 48 jam tidak menunjukkan perubahan nilai secara nyata (P>0,05) serta
masih berada dalam kisaran normal. Jumlah monosit cenderung mengalami
kenaikan dan secara statistik berbeda nyata pada penyimpanan 48 (P<0,05).
Morfometri jenis sel leukosit mengalami pembesaran diameter sebanding dengan
semakin lama penyimpanan. Selain perubahan ukuran sel, juga ditemukan
vakuolisasi inti dan sitoplasma pada monosit, neutrofil, dan eosinofil mulai 24 jam
penyimpanan. Complete blood count is one of the blood tests carried out to evaluate the
health of livestock. Some pre-analytical factors that also affect the data prodeced
include the type of anticoagulant used, the time lag of the examination, and the
storage temperature. Therefore, the factor is important and needs to be considered
before the blood examination is done. This study aimed to determine the number
and morphology of blood leukocytes given the Anticoagulant Tripotassium Ethylene
Diamine Tetraacetic Acid (K3EDTA) and stored for 48 hours at 4 °C. This study
were used 10 female ongole crossbred cattle (PO) cows aged 4 - 5 years. Blood
samples were collected through the jugular vein to observe the number and
morphology of leukocyte cell types with storage times of 0, 3, 6, 9, 12, 24, and 48
hours. The results showed that the total number of leukocytes, lymphocytes,
neutrophils, eosinophils, and basophils until 48-hour observation did not show a
noticeable change in values (P>0,05) and was still within the normal range. The
number of monocytes increase significantly at 48 hours of storage (P<0,05).
Morphometry of leukocyte cell types undergoes a magnification in diameter
proportional to the longer the storage. In addition to change in cell size,
vacuolization of nucleus and cytoplasm were also found in monocytes, neutrophils,
and eosinophils after being stored for 24 hours.
