Optimasi Imobilisasi Sefalosporin C Asilase Menggunakan Teknik Cross-Linked Enzyme Aggregates dengan Koagregat Kitosan

Sefalosporin merupakan antibiotik β-laktam semisintetik yang diprioritaskan oleh WHO dan sudah mencakup 30% dari total penggunaan antibiotik di dunia karena memiliki spektrum antibakteri yang luas dan lebih kebal terhadap enzim β-laktamase yang dihasilkan bakteri resisten antibiotik. Sefalosporin umumnya disintesis dari senyawa prekursor asam 7-aminosefalosporanat (7-ACA), hasil hidrolisis senyawa antibakteri alami sefalosporin C (CPC) yang dihasilkan jamur Acremonium chrysogenum. Dengan demikian, produksi 7-ACA yang efisien sangat penting dalam industri sefalosporin.
Produksi 7-ACA secara enzimatis terus dikembangkan karena lebih efisien dan ramah lingkungan dibandingkan proses kimia. Sefalosporin C asilase (CCA) merupakan enzim yang dapat mengkonversi CPC menjadi 7-ACA dalam satu tahap. Disisi lain, imobilisasi enzim diperlukan untuk menghasilkan biokatalis yang efisien untuk proses enzimatik skala industri. Imobilisasi terikat matriks umum dilakukan untuk keperluan tersebut karena terbukti dapat meningkatkan stabilitas enzim. Namun, penggunaan matriks menyebabkan penurunan aktivitas karena biokatalis mengalami dilusi enzim hingga 99%. Penggunaan matriks juga meningkatkan biaya produksi biokatalis dan menyisakan limbah matriks biokatalis nonaktif. Cross-linked enzyme aggregates (CLEA) merupakan teknik imobilisasi enzim tanpa matriks yang prospektif untuk mengatasi masalah tersebut. CLEA juga terbukti sebagai teknik yang sederhana dan efisien untuk mengimobilisasi enzim. Namun sampai saat ini, belum ada laporan mengenai aplikasi teknik CLEA untuk CCA.
Secara prinsip, teknik CLEA terdiri dari dua tahap utama, yaitu: presipitasi enzim bebas yang menghasilkan agregat enzim dan taut silang antar agregat enzim yang membentuk partikel CLEA. Dalam hal ini, presipitasi menggunakan amonium sulfat dan taut silang menggunakan glutaraldehida umum dilakukan. Namun, taut silang glutaraldehida kurang efektif ketika diaplikasikan pada enzim dengan sedikit residu asam amino lisin permukaan, enzim terimobilisasi mudah terlepas dari struktur CLEA saat pembilasan atau aplikasinya. CCA merupakan enzim heterodimer dengan sembilan (sedikit) residu lisin permukaan. CCA juga mengandung asam amino serin (dekat situs aktifnya) yang reaktif terhadap glutaraldehida sehingga berpotensi mengalami inaktivasi. Penambahan koagregat kaya gugus amino reaktif, seperti: bovin serum albumin, polietilenimin, atau polilisin dilaporkan menghasilkan partikel CLEA yang terikat kuat, dapat mencegah disosiasi subunit enzim, dan meminimalkan taut silang berlebihan glutaraldehida. Kitosan adalah koagregat alternatif yang kaya gugus amina reaktif, melimpah di alam, dan ramah lingkungan dibandingkan polimer atau biopolimer lain. Gugus amina reaktif kitosan potensial untuk memediasi taut silang agregat enzim. Oleh karena itu, Penambahan kitosan pada imobilisasi CCA-CLEA perlu dipertimbangkan. Penelitian ini bertujuan untuk mengoptimasi imobilisasi CCA-CLEA dengan koagregat kitosan agar menghasilkan enzim imobil yang memiliki aktivitas maksimum dan stabil terhadap perubahan pH dan suhu.
Dalam penelitian ini, lisat CCA dihasilkan dari fermentasi Escherichia coli BL21 (DE3)/ pET21a-S12 mutan yang disolasi melalui proses lisis sel dan sentrifugasi. Lisat CCA selanjutnya dimurnikan secara parsial menggunakan teknik salting-out dengan penambahan amonium sulfat jenuh 20-60% untuk menghasilkan suspensi fraksi aktif CCA. Suspensi tersebut kemudian dikoagregasikan dengan kitosan agar membentuk agregat CCA-kitosan. Agregat tersebut selanjutnya ditautsilangkan menggunakan glutaraldehida sehingga terbentuk partikel CCA-CLEA. Dalam penelitian ini, imobilisasi CCA-CLEA dioptimasi melalui tiga tahap, yaitu: (i) penapisan faktor signifikan menggunakan 2n-2 fractional factorial design (2n-2 FFD), (ii) pendugaan area optimum faktor signifikan menggunakan metode jalur pendakian tercuram, dan (iii) optimasi kondisi imobilisasi menggunakan response surface methodology (RSM) dengan central composite design (CCD). Aktivitas CCA-CLEA (U/g) digunakan sebagai respon. Stabilitas CCA-CLEA teroptimasi terhadap pH dan suhu juga dibandingkan dengan enzim bebas.
Eksperimen penapisan faktor signifikan menunjukkan bahwa konsentrasi glutaraldehida (%, v/v) dan waktu taut silang (menit) sebagai faktor signifikan dan empat faktor lainnya, konsentrasi kitosan (mg/ml), waktu koagregasi (menit), pH suspensi, dan suhu inkubasi (oC) tidak signifikan pada rentang yang diterapkan. Lebih lanjut, optimasi tahap 2 menghasilkan dugaan area optimum faktor signifikan, konsentrasi glutaraldehida 0,04-0,32% (v/v) dan waktu taut silang 118-120 menit, sementara faktor tidak signifikan dibuat konstan. Pada optimasi tahap 3, RSM dengan CCD menghasilkan model kuadrat yang memprediksikan aktivitas maksimum CCA-CLEA sebesar 86,02 U/g pada kondisi imobilisasi optimum, konsentrasi glutaraldehida 0,20% (v/v) dan waktu taut silang 119 menit. Eksperimen validasi menghasilkan aktivitas maksimum CCA-CLEA sebesar 85,91 U/g dan mengkonfirmasi bahwa model valid dengan galat relatif 0,13%. Selain itu, CCA-CLEA teroptimasi juga menunjukkan stabilitas yang lebih baik terhadap pH dan suhu dibandingkan CCA bebas.
Sebagai kesimpulan, optimasi imobilisasi CCA-CLEA dengan koagregat kitosan dapat menghasilkan aktivitas maksimum sebesar 85,91 U/g pada kondisi imobilisasi optimum, konsentrasi glutaraldehida 0,20% (v/v) dan waktu taut silang 119 menit. CCA-CLEA dengan koagregat kitosan juga memiliki stabilitas yang lebih baik terhadap pH dan suhu dibandingkan CCA bebas. Namun, CCA-CLEA masih memiliki beberapa kekurangan, yaitu: memiliki yield imobilisasi 60,31%, yield aktivitas 13,89%, dan mengalami dilusi enzim 44,85%. Oleh karena itu, pengembangan imobilisasi CCA-CLEA selanjutnya perlu mengeskplorasi faktor lain yang dapat mengatasi kekurangan tersebut atau mengembangkan strategi CLEA yang lebih mutakhir, seperti: CLEA berpori (p-CLEA). Cephalosporins are semisynthetic β-lactam antibiotics prioritized by WHO since have a broad antibacterial activity spectrum and are more stable against the β-lactamase produced by antibiotic-resistant bacteria. Currently, cephalosporins use covers 30% of total antibiotic use worldwide. Cephalosporins are generally synthesized from the precursor compounds 7-aminocephalosporanic acid (7-ACA), resulted from hydrolysis of natural antibacterial compounds cephalosporin C (CPC) produced by the fungal Acremonium chrysogenum. Therefore, it is essential to produce 7-ACA efficiently.
Currently, the enzymatic process for the 7-ACA synthesis continues to be developed since it is more efficient and environmentally friendly than chemical processes. Cephalosporin C acylase (CCA) is a potential enzyme because it can convert CPC to 7-ACA in one step. To produce efficient biocatalysts for industry, free enzymes should be immobilized. Matrix-bound enzyme immobilization has been proven to improve enzyme stability, but it decreases the activity due to enzyme dilution by up to 99%. Besides, the use of matrices increases the biocatalyst production cost and results in inactive biocatalyst matrix waste. Cross-linked enzyme aggregates (CLEAs) is a prospective matrix-less enzyme immobilization technique to address these problems. CLEA has been proven to be a simple, fast, and efficient technique for immobilizing various enzymes. However, there have been no reports regarding the CLEA technique application to CCA.
The CLEA technique involves two main steps: the precipitation of free enzymes to produce the enzyme aggregate and the crosslinking of the aggregate. In this case, precipitation using ammonium sulfate and aggregate crosslinking using glutaraldehyde are often used. However, glutaraldehyde crosslinking become less effective when applied to enzymes with few superficial lysine residues, the bound enzyme easily released from its CLEA structure. CCA is a heterodimeric enzyme with nine (slightly) superficial lysine residues. CCA also contains the amino acid serine (near its active site) which is reactive to glutaraldehyde so it is possible causing inactivation. The addition of coaggregates rich in reactive amino groups, such as bovine serum albumin, polyethylenemine, or polylysine has been reported to produce strongly bound CLEA particles, could prevent the enzyme subunits dissociation, and the excessive glutaraldehyde crosslinking. Chitosan is a alternative co-aggregate which is rich in reactive amino groups, more economic, abundant in nature, and environmentally friendly. The chitosan reactive amino group has the potential to mediate enzyme aggregates cross-linking. Therefore, the chitosan addition in the immobilization of CLEAs-CCA also needs to be considered. This study aimed to optimize the immobilization of CLEAs-CCA with chitosan as a co-aggregate.
In this study, CCA lysate was produced from the fermentation of mutant Escherichia coli BL21 (DE3)/ pET21a-S12 and then isolated through cell lysis and centrifugation processes. CCA lysate was partially purified using a salting-out technique with the addition of 20-60% saturated ammonium sulfate to produce a CCA suspension active fraction. Then, co-aggregation of the CCA active fraction suspension with chitosan was carried out to form the CCA-chitosan aggregates. The aggregates were then cross-linked with the addition of glutaraldehyde to form CLEAs-CCA. In this study, CLEAs-CCA immobilization was optimized through three stages, (i) screening for significant factors using 2n-2 fractional factorial design (2n-2 FFD), (ii) approximating the significant factor optimum area using the steepest ascent method, and (iii) optimization of immobilization conditions using response surface methodology (RSM) with central composite design (CCD). CLEAs-CCA activity (U/g) was used as a response. The stability of CLEAs-CCA against pH and temperature was also compared with free enzymes.
The screening significant factor experiments showed glutaraldehyde concentration (%, v/v) and cross-linking time (minutes) as significant factors and four other factors, chitosan concentration (mg/ml), co-aggregation time (minutes), suspension pH, and incubation temperature (oC) is not significant over the applied range. Furthermore, the approximation of the significant factors optimum area resulted in an area of glutaraldehyde concentration of 0.04-0.32% (v/v) and cross-linking time of 118-120 minutes, while insignificant factors are kept constant. Finally, RSM with CCD produces a quadratic model that predicts the maximum activity of the CCA-CLEA 86.02 U/g obtained under optimum immobilization conditions, 0.20% glutaraldehyde concentration (v/v) and 119 minutes of cross-linking time. The validation experiment resulted in a maximum CCA-CLEA activity of 85.91 U/g and confirmed that the model was valid with a relative error of 0.13%. In addition, CLEAs-CCA also has better pH and temperature stability than free CCA.
In conclusion, optimization of CLEAs-CCA immobilization with chitosan coaggregate produce maximum activity of 85.91 U/g at optimum immobilization conditions, glutaraldehyde concentration 0.20% (v/v) and cross-linking time of 119 minutes. CLEAs-CCA with chitosan coaggregate also has better pH and temperature stability than free CCA. However, CLEAs-CCA still has several drawbacks, it has an immobilization yield of 60.31%, an activity yield of 13.89%, and has enzyme dilution of 44.85%.Therefore, the development of CLEAs-CCA still needs exploring other factors to address the problems or developing the latest CLEAs strategy, such as porous-CLEAs (p-CLEAs).
