dc.description.abstract | Cumi-cumi merupakan moluska tanpa cangkang yang termasuk dalam ordo Chepalopoda dan merupakan komoditas yang bernilai ekonomi penting, pemanfaatannya yang terus mengalami peningkatan menyebabkan perlunya studi untuk menjaga kelestariannya. Penentuan spesies cumi-cumi secara morfologis tidak cukup akurat sehingga teknik identifkasi biomolekuler lebih baik digunakan dalam pengelolaan. Penelitian ini bertujuan untuk mengautentikasi spesies cumi-cumi yang dieksploitasi di perairan Belawan melalui teknik biomolekuler, periode penelitian antara Januari 2020 hingga Agustus 2020 meliputi studi lapang, pengambilan contoh, pengamatan morfologi dan DNA barcoding. Selanjutnya dilakukan proses preparasi, ekstraksi, amplifikasi, dan PCR, sequencing yang kemudian diolah data menggunakan MEGA X. Cumi-cumi dari perairan Belawan diidentifikasi sebagai Uroteuthis chinensis yang memiliki nama umum Mitre squid kemudian divalidasi menggunakan biomarka gen COI dengan nilai 99,78%. dc.description.abstract | Squids are molluscs without shells belong to order Cephalopoda which are economically important commodities, leads to high concern of its sustainibility amid an ever increasing utilization. Species determination in squids may be insufficient morphologically, instead biomolecular techniques are used for better management. The present study aims to authenticuate squids species being exploited in Belawan waters through biomolecular technique, research period was between January 2020 to August 2020 covering fieldwork, sample collection, morphological examination and DNA barcoding on squids. In the following of preparation, extraction, amplification, and PCR processes, sample was sequenced and processed using MEGA X. The squid obtained from Belawan waters was identified as Uroteuthis chinensis, which commonly named Mitre squid and then was validated using the COI gene biomarker with a value of 99,78%.
