      Induksi Proliferasi Tunas Grammatophyllum stapeliiflorum J.J.Sm pada Berbagai Komposisi Media dan Benzyl Adenin secara In Vitro

      Abstract
      Anggrek Grammatophyllum stapeliiflorum J.J.Smith termasuk kedalam kelompok Appendix II. Perbanyakan secara konvensional sulit untuk memenuhi kebutuhan bibit yang relatif cepat, sehingga dilakukan perbanyakan secara in vitro untuk mempersingkat perbanyakan tanaman dan memiliki kualitas yang baik. Penelitian bertujuan memperlajari daya pembentukan tunas Grammatophyllum stapeliiflorum pada beberapa komposisi dan konsentrasi media serta penambahan BA secara in vitro. Percobaan ini disusun menggunakan Rancangan Kelompok Lengkap Teracak faktorial tiga faktor dengan bahan tanam Grammatophyllum stapeliiflorum aksesi Sulawesi. Faktor pertama adalah komposisi media: Knudson C dan Schenk and Hildebrandt; faktor kedua adalah konsentrasi hara makro dan mikro: ½, ¾, 1; faktor ketiga adalah konsentrasi BA: 0, 1, 2, 3 mg l -1 . Hasil percobaan menunjukan bahwa media terbaik untuk menginduksi tunas adalah media ½ SH + 2 mg l -1 BA menghasilkan rata-rata 7,75 tunas dalam 10 Minggu Setelah Tanam. Komposisi media yang optimal untuk pembentukan planlet adalah media ½ SH + tanpa BA dengan jumlah 35 planlet yang terbentuk.
       
      Grammatophyllum stapeliiflorum J.J.Smith is included in the Appendix II lists species. Conventional propagation is difficult to meet the needs of relatively fast seedlings, so it can be done with in vitro propagation to have a good quality of seedlings in a short time. The aim of this research was to find the best combination of medium composition and concentration with an addition of BA for the proliferation of shoots formation. This experiment was designed using a factorial Randomized Complete Block Design with three factors. The first factor is the type of media composition: Knudson C, Schenk and Hildebrandt; the second factor is macro and micronutrient concentration: ½, ¾, 1 level; the third factor is BA concentration: 0, 1, 2, 3 mg l -1 . The experimental results showed that the optimum medium to induce shoot proliferation in this study was ½ SH + 2 mg l -1 BA medium produced an average of 7.75 shoots per explant. The optimum medium to induce plantlet was ½ SH + 0 mg l-1 BA medium that produced 35 plantlets.
