      Study of Senna siamea Leaf Extract as Botanical Insecticide Candidate against Spodoptera litura

      Abstract
      Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) is a polyphagous insect, well-known as a major pest in many countries due to its potency to cause failure of many important crops. Efforts to control this pest using synthetic insecticides end up with residues contaminating soil and water and intoxicate the agriculture products. Previous studies showed that Senna siamea has insecticidal properties and affects the activity of detoxification enzymes against pest insects. This research is aimed to study the insecticidal effects of S. siamea extract against S. litura by a toxicity assay and detoxification enzyme assay. S. siamea leaves were extracted using hexane, dichloromethane, ethyl acetate, and methanol as solvents separately. The extract was then tested againts the second instar larvae of S. litura. The results showed that all the extracts had toxicity effects against S. litura with the lowest LD50 of 7.254 µg/larva after 24 hours of treatment by methanol extract. The extracts also affected the detoxification enzyme activities by inhibiting carboxylesterase activities while inducing glutathione S-transferase and acetylcholinesterase activities. Although S. siamea leaf extracts had acute toxic effect on S. litura, the extracts also increased their enzymatic activities that lead to possible toxin resistance in long term use.
       
      Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) merupakan serangga polifagus, yang dikenal sebagai hama utama karena potensinya menyebabkan gagal panen pada beberapa produk pertanian di banyak negara. Upaya pengendalian hama menggunakan insektisida sintetis dapat menyebabkan kontaminasi berupa residu di tanah, air, dan produk pertanian. Hasil penelitian terdahulu menunjukkan bahwa Senna siamea terbukti memiliki sifat insektisida dan memberikan efek pada aktivitas enzim detoksifikasi. Penelitian ini bertujuan mempelajari efek insektisida ekstrak S. siamea terhadap S. litura dengan uji toksisitas dan uji enzim detoksifikasi. Penelitian ini dilakukan dengan mengekstraksi daun S. siamea kering dengan hexana, dichloromethane, etil asetat, dan metanol sebagai pelarut. Setiap ekstrak kemudian diujikan pada instar kedua larva S. litura. Hasil penelitiaan ini menunjukkan bahwa seluruh ekstrak tersebut memiliki efek toksisitas terhadap S. litura dengan LD50 terendah 7.254 µg/larva setelah 24 jam perlakuan ektrak metanol. Ekstrak ini juga mempengaruhi aktivitas enzim detoksifikasi dengan menghambat aktivitas karboksilesterase, tetapi menginduksi aktivitas glutathioneS-transferase dan asetilkolinesterase. Meskipun ekstrak S. siamea memiliki efek toksik yang akut terhadap S. litura, ekstrak ini juga meningkatkan aktivitas enzimatik yang mengarah pada resistensi apabila digunakan pada jangka panjang.
