MAERA ZASARI. Studi Toleransi Benih Propelegitim Kakao terhadap Cekaman
Kekeringan. Dibimbing oleh SUDARSONO sebagai ketua, ADE WACHJAR dan
AGUNG WAHYU SUSILO sebagai anggota komisi pembimbing.
Kondisi kekeringan merupakan ancaman bagi budidaya tanaman kakao,
karena mengakibatkan kematian benih, menurunkan produksi, meningkatkan
serangan hama dan penyakit, serta degradasi lahan. Benih kakao yang toleran
dibutuhkan untuk mengatasi permasalahan akibat cekaman kekeringan. Keberadaan batang bawah yang toleran kekeringan sangat bermanfaat dalam
pemuliaan benih kakao, karena peranannya yang berhubungan langsung dengan
kondisi defisit air pada lingkungan tumbuh tanaman. Batang bawah kakao dapat
diproduksi dari perbanyakan biji propelegitim yang tersedia banyak. Benih asal
biji propelegitim menjadi alternatif untuk batang bawah kakao yang toleran
kekeringan.
Tujuan umum penelitian adalah untuk mendapatkan genotipe benih
propelegitim kakao harapan yang bersifat toleran kekeringan untuk digunakan
sebagai batang bawah kakao. Tujuan khusus penelitian ini adalah untuk: (1)
mengidentifikasi toleransi kekeringan benih propelegitim terhadap cekaman
kekeringan, (2) mengidentifikasi karakter fisiologi dari genotipe benih
propelegitem pada kondisi kekeringan, (3) mengidentifikasi toleransi biji
propelegitim terhadap cekaman kekeringan, dan (4) mengidentifikasi sifat toleran
kekeringam berbasis marka spesifik asal gen DREB2.
Materi genetik koleksi Pusat Penelitian Kopi dan Kakao Indonesia
(Puslitkoka) yang terdiri dari 13 klon kakao lindak, yaitu KW 516, KW 617, KW
641, ICCRI 03, Scavina 06, TSH 858, KEE 02, KKM 22, ICS 60, MCC 02,
Sulawesi 01, Sulawesi 02, dan Sulawesi 03. Materi genetik merupakan klon-klon
unggulan yang diantaranya yaitu KW 516, KW 641, Sulawesi 01, Sulawesi 03,
dan KKM 22 terindikasi bersifat toleran, sedangkan ICS 60 tergolong peka.
Hasil seleksi toleransi kekeringan benih yang ditumbuhkan pada media
tanam berkadar lengas air 25 %, mendapatkan bobot basah akar sebagai kriteria
untuk membedakan sifat toleransi kekeringan benih. Analisis indek sensitivitas
kekeringan (ISK) dan penurunan relatif (PR) terhadap karakter seleksi
mendapatkan 2 tipe sifat toleransi kekeringan pada genotipe benih, yaitu: (1)
genotipe bersifat toleran moderat meliputi genotipe benih asal klon KKM 22,
ICCRI 03, ICS 60, TSH 858, MCC 02, dan KW 641; dan (2) genotipe bersifat
peka meliputi genotipe benih asal klon Scavina 06, KW 516, Sulawesi 01, KEE
02, Sulawesi 03, KW 617, dan Sulawesi 02. Hasil identifikasi terhadap kandungan air relatif, densitas stomata, dan
kandungan prolin dari genotipe benih propelegitim yang mengalami cekaman
kekeringan, mendapatkan; (1) cekaman kekeringan menyebabkan perubahan
signifikan terhadap kandungan air relatif dan kandungan prolin, tetapi tidak
mempengaruhi densitas stomata pada genotipe benih, (2) Sebagian genotipe benih
bersifat toleran mengalami penurunan kandungan air relatif dan kenaikan prolin
yang signifikan berbeda terhadap genotipe benih bersifat peka, sedangkan penurunan densitas stomata antara kedua kelompok benih cenderung tidak
berbeda.
Evaluasi sifat toleransi kekeringan pada stadia perkecambahan dalam media
yang diinduksi larutan PEG, menjadikan karakter daya berkecambah sebagai
kriteria seleksi untuk menentukan toleransi kekeringan dari genotipe biji. Analisis
indek senstivitas kekeringan (ISK) dan penurunan relatif (PR) karakter daya
berkecambah didapatkan 6 (enam) genotipe biji bersifat toleran moderat, yaitu
genotipe biji asal klon Scavina 06, KW 641, ICCRI 03, KW 617, TSH 858, dan
Sulawesi 02; dan 7 (tujuh) genotipe biji bersifat peka, yaitu genotipe biji asal klon
Sulawesi 03, KEE 02, KKM 22, ICS 60, MCC 02, KW 516, dan Sulawesi 01. Pendugaan sifat toleransi kekeringan klon dan genotipe benih berbasis
marka gen DREB2 menunjukkan: (1) 12 klon selain TSH 858 memiliki gen
DREB2 atau terindikasi bersifat toleran, (2) benih yang memiliki gen DREB2 atau
terindikasi bersifat toleran antara lain genotipe benih asal KW 617, Sulawesi 03,
Sulawesi 01, KW 516, KKM 22, dan ICCRI 03, dan (3) sekuensing terhadap
DNA DREB2 asal klon mendapatkan sejumah kecil perubahan pada runutan basa
nukleotida DNA_F (forward) dan DNA_R (reverse). Perubahan basa nukleotida
pada tiap-tiap sekuen terindikasi akibat mutasi dari proses substitusi, tranversi,
delesi, dan insersi. Analisis kemiripan tipe mutasi dan jumlah perubahan basa
mendapatkan 11 sekuen DNA DREB2 asal klon tetua yang memiliki tingat
kemiripan 72.26 %, sedangkan sekuen DNA DREB2 asal klon Scavina 06
memiliki tingkat kemiripan paling rendah. Mutasi pada basa nukleotida
menghasilkan keragaman pada sekeun DNA_F dan DNA_R asal klon tetua,
sebagaimana yang ditunjukkan oleh pengelompokkan sekeun DNA DREB2 pada
hasil analisis filogenetik . Berdasarkan hasi studi didapatkan hasil inkonsistensi pada sifat toleran
kekeringan. Terdapat genotipe yang bersifat toleran pada fase benih menunjukkan
sifat peka pada fase biji atau berdasarkan marka gen DREB2. Inkonsistesi sifat
toleransi kekeringan diduga karena keragaman sifat ketahanan yang diwarisi oleh
genotipe progeni (benih atau biji) propelegitim dari klon tetua yang bersari bebas.
Klon tetua yang bersilang acak memungkinkan terjadinya proses rekombinasi
genetik yang akan diwariskan pada keturunannya (progeni). Sifat atau karakter
tanaman tidak dikendalikan oleh satu gen, melainkan setiap gen berinteraksi
dengan gen lainnya dalam membentuk fenotipe.

Drought conditions are a threat to cocoa cultivation, because they cause the
death of seedling, decreasing production, increasing pest and disease, and
degrading the land. Tolerant cocoa seedlings are needed to overcome problems
caused by drought stress. The existence of drought-resistant rootstocks is very
useful in breeding the tolerant cocoa seedlings because this is directly related to
water deficit conditions in the plant's growing environment. Cocoa rootstock can
be produced from the propagation of widely available prope legitimate seeds. The
prope legitimate seeds are an alternative for drought-tolerant cocoa rootstock.
The general objective of the study was to obtain the drought tolerance
genotypes of the expected cocoa prope legitimate for rootstock. The specific
objectives of this study were to: (1) identify the drought tolerance of prope
legitimate seedlings to drought stress, (2) identify the physiological characteristics
of the prope legitimate seedling genotypes under drought conditions, (3) identify
the tolerance of the prope legitimate seeds to drought stress, and (4) identify
drought tolerant based on the DREB2 gene marker. The genetic material collected by the Indonesian Coffee and Cocoa
Research Center (Puslitkoka) consisting of 13 cocoa clones, namely KW 516, KW
617, KW 641, ICCRI 03, Scavina 06, TSH 858, KEE 02, KKM 22, ICS 60, MCC
02, Sulawesi 01, Sulawesi 02, and Sulawesi 03. Genetic material is superior
clones, including KW 516, KW 641, Sulawesi 01, Sulawesi 03, and KKM 22
indicated as tolerant, while ICS 60 is classified as sensitive. The results of the drought tolerance selection for seedlings grown on a 25%
moisture content planting medium obtained the root wet weight as a criterion to
differentiate the drought tolerance properties of seeds. Analysis of the drought
susceptibility index (DSI) and relative reduction (RR) on the character selection
obtained 2 types of drought tolerance traits in the seed genotype, namely: (1) the
genotype was moderately tolerant including the genotype of seeds from clone
KKM 22, ICCRI 03, ICS 60, TSH 858, MCC 02, and KW 641; and (2) sensitive
genotypes include seed genotypes from clones of Scavina 06, KW 516, Sulawesi
01, KEE 02, Sulawesi 03, KW 617, and Sulawesi 02.
The identification results of the relative water content, stomatal density, and
proline content of the prope legitimate seedling genotypes experiencing drought
stress, obtained; (1) drought stress caused significant changes in relative water
content and proline content, but did not affect stomatal density in seedling
genotypes, (2) Some of the tolerance genotypes were showed a decrease in
relative water content and a significantly different increase in a proline to the
sensitive genotypes, whereas the decrease in stomata density between the two
groups of seedling genotypes were not different.
The evaluation of drought tolerance for the germination stage in media
induced by PEG solution, obtained the character of germination capacity as a selection criterion to determine drought tolerance of seed genotypes. Analysis of
drought susceptibility index (DSI) and relative reduction (RR) of germination
the character showed that 6 (six) of the seed genotypes were moderately tolerant,
namely the genotypes from clones of Scavina 06, KW 641, ICCRI 03, KW 617,
TSH 858, and Sulawesi 02; and 7 (seven) of the sensitive seed genotypes, namely
the genotypes from clones of Sulawesi 03, KEE 02, KKM 22, ICS 60, MCC 02,
KW 516, and Sulawesi 01.
The estimation of the drought tolerance of clones and seedlings based on
DREB2 gene markers showed: (1) 12 clones other than TSH 858 had DREB2
genes or indicated to be tolerant, (2) the seedlings that had DREB2 genes or
indicated to be tolerant, among others, genotypes of seeds from KW 617,
Sulawesi 03, Sulawesi 01, KW 516, KKM 22, and ICCRI 03, and (3) sequencing
of DREB2 DNA from clones obtained a small number of changes in the DNA_F
(forward) and DNA_R (reverse) nucleotide base sequences. Changes in nucleotide
bases in each sequence were indicated as the result of mutations from the
substitution, transversion, deletion, and insertion processes. Analysis of the
the similarity of mutation type and number of base changes obtained the eleven (11)
DREB2 DNA sequences from parent clones which had a similarity level of
72.26%, while DREB2 DNA sequences from Scavina 06 clone had the lowest
similarity level. The mutations of the nucleotide bases resulted in variations of the
DNA_F and DNA_R sequences from the parent clones, as shown by DREB2
DNA sequence grouping in the results of the phylogenetic analysis.
Based on the results of the study, inconsistency results were obtained from the
drought-tolerant. There are genotypes that are tolerant at the seedling stage
showing sensitivity in the seed stage or based on DREB2 gene markers. The
inconsistency of the drought-tolerant is thought to be due to the diversity of the
resistance traits inherited by the progeny genotype (seedlings or seeds) of the
prope legitimate from randomly crossed parental clones. The parental clones that
cross randomly allow the genetic recombination process to be inherited from the
offspring/progeny. The characters are not controlled by one gene, but each gene
interacts with other genes to form phenotypes.
