      Kepastian Spesies Macrobrachium sintangense (de Man, 1898) Menggunakan Gen Parsial COI dari Perairan Danau Lido, Jawa Barat

      Abstract
      Macrobrachium sintangense dengan nama lokal udang regang pernah diinformasikan bahwa tidak ditemukan pada periode Desember 2015-Mei 2017 di perairan Danau Lido, Jawa Barat. Oleh karena itu, kepastian spesies udang yang teridentifikasi secara morfologi sebagai M. sintangense dibutuhkan untuk memperbaiki pengelolaan sumberdaya. Penelitian ini bertujuan untuk mengidentifikasi dan memastikan spesies udang regang menggunakan pendekatan molekuler dengan marka gen parsial COI. DNA total diisolasi dari jaringan sampel menggunakan kit komersil (Geneaid). Tiga DNA sampel berkualitas baik diperoleh pada tahap amplifikasi untuk disequencing. Hasil sequencing disejajarkan menggunakan MEGA X. Hasil BLASTn diperoleh bahwa sekuen sampel lebih mirip dengan sekuen Macrobrachium sp. AB235301.1 sebesar 95% dengan 26 situs basa nukleotida spesifik. Sementara itu, sekuen M. sintangense MF622027.1 ditemukan pada database gen COI. Analisis filogeni menunjukkan bahwa sekuen sampel berada pada posisi clade terpisah dengan sekuen database M. sintangense. Sampel udang yang teridentifikasi secara morfologi sebagai M. sintangense belum tervalidasi dengan pendekatan molekuler dengan marka gen parsial COI sebagai spesies yang sama.
       
      Macrobrachium sintangense with the local name regang shrimp has been informed that it was not found in the period December 2015-May 2017 in the Lake Lido waters, West Java. Therefore, certainty of the species of shrimp identified morphologically as M. sintangense is needed to improve resource management. This study was aimed to identify and confirm the species of regang shrimp using a molecular approach with COI partial gene markers. Total DNA was isolated from the tissue sample using commercial kit (Geneaid). Three good quality DNA samples were obtained at the amplification stage for sequencing. Sequencing results were aligned using MEGA X. The BLASTn results showed that the sample sequence most similar to the Macrobrachium sp. sequence by 95% with 26 specific nucleotides. Meanwhile, M. sintangense MF622027.1 sequence was found in the COI gene database. Phylogeny analysis showed that the samples sequences were in a separate clade position with the M. sintangense database sequence. Shrimp samples identified morphologically as M. sintangense have not been validated by a molecular approach with the COI partial gene markers as the same species.
