      Deteksi Bakteri Pembentuk Amina Biogenik pada Ikan Scombridae Melaui Gen Penyandi dan Primer Spesifik Secara Multiplex-PCR

      Abstract
      Amina biogenik merupakan komponen basa nitrogen yang terbentuk oleh dekarboksilasi asam amino. Amina biogenik yang sering terdeteksi pada produk perikanan di antaranya histamin, tiramin dan kadaverin yang dihasilkan oleh bakteri. Deteksi bakteri dapat dilakukan melalui gen penyandi yang dihasilkan bakteri pembentuk amina biogenik serta deteksi melalui spesifik bakteri pembentuknya. Gen pengkode dekarboksilasi asam amino tersebut yaitu histidine decarboxylase (hdc), tyrosine decarboxylase (tdc) serta lysine decarboxylase (ldc). Penelitian ini bertujuan untuk mendeteksi gen hdc, tdc dan ldc menggunakan multiplex PCR dan mengidentifikasi bakteri pembentuk amina biogenik pada ikan scombridae. Sampel yang digunakan yaitu ikan tuna beku, tongkol beku, cakalang beku, tuna loin, pindang potong dan pindang bumbu kuning. Ikan tuna yang disimpan pada suhu ruang selama 3 hari sebagai kontrol positif, kontrol negatif berupa ddH 2 O dan media enrichment. Penelitian terdiri dari enrichment bakteri pada media lactose broth dan marine broth, isolasi DNA sampel daging ikan dan hasil enrichment, uji kemurnian dan konsentrasi isolat, amplifikasi gen target hdc, tdc, ldc menggunakan multiplex PCR, sekuensing, analisis bioinformatika bakteri pembentuk amina biogenik, desain primer dan amplifikasi spesifik bakteri Morganella morganii dan Carnobacterium divergens sebagai pembentuk amina biogenik menggunakan multiplex PCR. Hasil yang diperoleh yaitu isolat DNA bakteri dengan rasio kemurnian (A260/280) yaitu 1,49-2,17 serta konsentrasi isolat 15,75-461,75 ng/µL. Marine broth merupakan media enrichment terpilih yang dapat mengamplifikasi 3 gen target hdc, tdc dan ldc menggunakan multiplex PCR. Gen tdc, hdc dan ldc teramplifikasi menggunakan multiplex PCR pada sampel tuna beku, pindang potong dan pindnag bumbu kuning. Bakteri pembentuk amina biogenik histamin (gen hdc) yang teridentifikasi pada sampel tuna beku dan pindang potong yaitu Acinetobacter baumannii sedangkan pada pindang bumbu kuning yaitu Morganella morganii. Bakteri pembentuk tiramin (gen tdc) pada tuna beku, pindang potong dan pindang bumbu kuning yaitu Enterobacter cloacae. Bakteri pembentuk kadaverin (gen ldc) pada tuna beku dan pindang bumbu kuning teridentifikasi Enterobacter cloacae sedangkan pada pindang potong yaitu Enterobacter hormaechei. Primer spesifik bakteri M. morganii dan C. divergens sebagai pembentuk amina biogenik ditemukan pada seluruh sampel ditandai dengan teramplifikasi menggunakan multiplex PCR.
       
      Biogenic amines are nitrogenous base components formed by amino acid decarboxylation. Biogenic amines that are often detected in fishery products are histamine, tyramine and cadaverine which are produced by bacteria. Bacterial detection can be carried out by coding genes and specific forming bacteria. The amino acid decarboxylation coding genes are histidine decarboxylase (hdc), tyrosine decarboxylase (tdc) and lysine decarboxylase (ldc). The purpose of this study aims to detection hdc, tdc and ldc gene using multiplex PCR and identification of biogenic amine producing bacteria in scombridae fish. The specimens used were frozen tuna, little tuna, skipjack and loin tuna. In addition, Indonesian traditional tuna such as pindang potong, pindang bumbu were observed. Tuna stored at room temperature for 3 days as positive control, ddH2O, medium pre-enrichment as control negative. The study consisted of the preenrichment stage of bacteria on lactose broth and marine broth medias, DNA isolation (sample with and non pre-enrichment), purity test and isolate concentration of target gene of hdc, tdc, ldc using multiplex PCR, sequencing, bioinformatics analysis of biogenic amine forming bacteria, desain and detection Morganella morganii and Carnobacterium divergens as histamine and tyramine forming bacteria. The result obtained were generally in a good quality of bacterial DNA isolates, ratio A260/280: 1.49-2.17 with concentration ranged between 15.75-157.84 ng/μL. Marine broth is a selected enrichment medium that can amplify hdc, tdc and ldc using multiplex PCR. The tdc, hdc and ldc genes were amplified using multiplex PCR on samples of frozen tuna, pindang potong and pindang bumbu. The histamine forming bacteria identified in frozen tuna and pindang potong was Acinetobacter baumannii, while on pindang bumbu was M. morganii. Tyramine forming bacteria in frozen tuna, pindang potong and pindang bumbu is Enterobacter cloacae. Cadaverin forming bacteria in frozen tuna and pindang bumbu is Enterobacter cloacae, while those on pindang potong is Enterobacter hormaechei. Specific primer M. morganii and C. divergens successfully amplified on all samples using multiplex PCR.
