dc.description.abstractThis research was designed in four experiments to determine the effectiveness of supplementation of various types and concenbations of sera and cocultures of Fallopian tube epithelial and cumulus cells in Tissue Cutture Medium 199 (TCM-199) on in vitro bovine embryo production. In the first experiment oocytes were collected by aspiration method from ovaries obtained from the slaughter house. Oocytes were classified into three grades (A, B and CID) according to their qualities. The second experiment dealt with maturation of cmcytes of A and B qualities (COCs, cumulus oocyte complexes) in TCM-199 supplemented with 5, 10, 15 and 20 percents of fetal bovine serum (FBS), estrous heifer serum (SHg) and men days after estrous heifer serum (SH7), respectively, at 38.5°C. 5.0% Cq2 in humidified air for 22 to 24 hours. Oocytes were thea fedid in vitro with capcitated spematozoa. After six to eight hours of culturing, the presumptive zygotes were tiansferred into 100 pl drop of in virro culture medium for development of embryos. These embryos were then incubated for 4x48 hours, evaluated and its culture medium was changed with a fresh one every 48 hours.
