Staphylococcus aureus is one of the main bacteria that causes subclinical mastitis in dairy cow. The most important virulent factor in early infection of mastitis in dairy cow is the existence of protein A of S. aureus. The objective of this research is to observe the characteristics and existence of S. aureus in milk sample from dairy cows that suffer from subclinical mastitis. The characterization of S. aureus are the ability to ferment mannitol, the catalase and coagulase activity, and the positive reaction to glucose test. The existence of protein A in S. aureus is determined by the serum soft agar (SSA) method. The serum used in this study is obtained from rabbit, sheep, and chicken sera. Fourteen isolates identified as S. aureus are tested with the SSA technique. Based on the SSA examination result, three isolates are reported having the ability to produce protein A. This also indicates that not all of S. aureus strain have the ability to produce protein A on their cell wall.id
