Karakteristik Semen Segar, Morfologi, dan Pengujian Keutuhan Membran Plasma Spermatozoa Kelinci Lop dan Rex

Abstract

The study was conducted to investigate the quality of semen between Lop and Rex breed. Semen samples was collected by using artificial vagina and evaluated macro and microscopically. The evaluation included semen volume, pH, consistency, color, mass movement, motility, individual scores, concentration, and viability. The sperm morphology was examination using carbofuchsin eosin staining and the plasma membran integrity was determine using Hypo-Osmotic Swelling test (HOS test). Hypo-Osmotic solution was modified by mixing fructose and sodium citrate into three different osmotic pressure: 50, 100, and 150 mOsm/kg. All data were analyzed statistically using analysis of variance (ANOVA), whereas the data from HOS test were analyzed using completely randomized design (CRD) with three factor patterns. The result showed that mass movement, motility, and individual scores of Rex’s sperm was superior compare than Lop’s, but there were no difference in other parameters. The respons of plasma membrane integrity showed that Rex’s sperm was faster (15 minute) compare than Lop’s (30 minute). This fact may relate to the composition structure of the plasma membrane and seminal plasma.
