This research was designed to understand utilization of embryonated egg as cultivation media and infection model of parasite Leucocytozoon caulleryi. An examination included percentage of parasitemia and leukocyte differential of chicken embryo. This research used 24 embryonated egg which were inoculated with gametocytes of L. caulleryi via chorioallantoic membrane (CAM) and allantoic cavity (AC) route. Inoculation were done at 13 days old and blood smear of embryo were done at 16, 17,18, and 19 days old. The result showed significant different of inoculation route and each embryonated egg’s age. Inoculation route via CAM showed higher parasitemia than AC. The highest of parasitemia were showed at 19 days old of embryonated egg (6 days post-inoculation) both CAM and AC. Percentage of leukocyte differential showed significant different of both inoculation route and embryo’s egg on 16, 17, 18, and 19 days old. Both CAM and AC route showed same percentage of lymphocyte as the highest percentage followed by heterophil, monocyte, basophil, and eosinophil. Thus, embryonated egg could be used as alternative cultivation media with chorioallantoic membrane as better inoculation route and infection model which was showed by imune response from leukocyte differential.en
