dc.description.abstractThe Red Honey Bee, Apis koschevnikovi was distributed in Semenanjung Malaysia, Sumatera, Java and Borneo. These species was found easily in Borneo but was rare in other place. Nearly all research on this species has been conducted at one site (Tenom, Malaysia) where it is common. Thus, basic research considering to distribution, ecology and genetic diversity need to conduct attemp to . this bee species in @.theast Kalimantan. Distribution maps could shows the original di,stribution or nattJal site where . these spesies could found at recent time. Ekologi shows natural hah.i..a t and spesific condition that could be basic of beekeeping management. While, III gen!ic diversity associated with viabilities and adaptation to environtment. Beside, genQic diversity also usefull to recognize fenotipes of A. koschevnikovi at spesific site"~ s basic rearing and breeding of these bee spesies. In order to describe more ::::I predisely recent distribution of this species in Southeast Kalimantan, a survey, r+ c:: samiJing methode and ecological observation was conduct attemp to this bee· CD spe~s. While, in order to evaluate the genetic diversity of this spesies, an analysis ::::I usinl RFLP is held in laboratory. E2 and HI primer was used to amplify intergenic tll re~ COX I-II, while Cyt b forward and reverse primer was used to amplify Cyt b. o - Intergenic region COX I-II was cut by Mbol (COX I-II/MboI) and SspI (COX 1- II1SspI), while Cyt b was cut by MboI (Cyt b / Mbol) and DraI (Cyt b / DraI). As the result, COXI-II/MboI shows one haplotipe, and COXI-II/Sspl shows two haplotipe. In Cyt b gene, Cyt b / MboI shows two haplotipe, and Cyt b / DraI shows one haplotipe.en
dc.description.abstractLebah madu Apis koschevnikovi terdistribusi di Semenanjung Malaysia, Sumatera, Jawa dan Borneo. Spesies ini mudah di temukan di Borneo tetapi di temp at lain jarang ditemukan. Hampir seluruh penelitian tentang spesies ini dilakukan di Tenom, Malaysia di mana spesies ini umum ditemukan. Oleh karena itu penelitian dasar mengenai distribusi, ekologi dan keragaman genetik spesies lebah ini di @ limantan Selatan perlu dilakukan. Pola distribusi dapat menggambarkan pen~baran asH dan lokasi alami dimana dapat ditemukan lebah Apis koschevnikovi saat.a.. ni. Ekologi dapat menggambarkan habitat alami dan kondisi spesifik sehingga !II dap~. menjadi dasar pengembangan budidaya lebah A. koschevnikovi. Sedangkan kera~aman genetik penting untuk diketahui karena erat kaitannya dengan viabilitas dan"~ emampuan adaptasi. Selain itu keragaman genetik juga bermanfaat untuk ::::I men.~.. usuri sifat-sifat genetik lebah A. koschevnikovi sebagai dasar budidaya dan r:::: bree~g lebah ini. Untuk memahami pola distribusi lebah A. koschevnikovi terkini di ~ . Kali;lantan Selatan, dilakukan survey, pengambilan contoh dan pengamatan ekologi. ::::I Se~gkan untuk mempelajari keragaman genetik, dilakukan analisis RFLP di laboitorium. Primer (Oligonukleotida) E2 dan HI digunakan untuk mengamplifIkasi o . daerab intergenik SO I-II. Sedangkan primer Sit b Forward dan reverse digunakan untuk mengamplifIkasi gen Sit b. Daerah intergenik SO I-IT dipotong dengan MboI (SO I-IUMboI) dan SspI (SO I-II/SspI). Sementara itu gen Sit b dipotong dengan MboI (Sit b/MboI) dan Dral (Sit b/DraI). HasH yang diperoleh, SO I-II/MboI menghasilkan satu haplotipe, SO I-II/SspI menghasilkan dua haplotipe. Sit b/MboI menghasilkan dua haplotipe sedangkan Sit b/DraI menghasilkan satu haplotipe.
