Production of Yolk Immunoglobulin Anti-Excretory/Secretory (E/S) of Fasciola gigantica to Laying Hens

dc.description.abstractThe research was designed to study the method of antibody antiexcretory/ secretory (E/S) of Fasciola gigantica production in laying hens. Ten chickens were divided into two groups, i.e., five chickens/group. Group one was immunized with antigen E/S of Fasciola gigantica from buffalo and the second group was immunized with antigen E/S of Fasciola gigantica from sheep. Dose of immunization was 150 μg/animal. The immunization was done intravenously (i.v.) without adjuvant at the first time. One week later (second immunization) all chicken was injected subcutaneously (s.c.) with the antigen that had been added with Freund’s complete adjuvant with the same volume (1:1). Since the third immunization until the sixth immunization whereas every immunization was conducted one week after the previous one, the chicken was injected subcutaneously with combination of antigen and Freund’s incomplete adjuvant. The eggs were collected every day after the first immunization. Sera and egg yolk were tested using agar gel precipitation test (AGPT) to detect the presence of antibody against antigen E/S of Fasciola gigantica. The result showed that the chicken immunized with antigen-E/S of Fasciola gigantica from buffalo was able to develop antibody aster than the chicken immunized with antigen-E/S of Fasciola gigantica from sheep. The antibody against antigen E/S of Fasciola gigantica antigen from buffalo appeared in the sera at the 9-week and in the egg yolk at the 10-week after the first immunization. While the antibody against antigen E/S of Fasciola gigantica from sheep appeared in the sera at the 10-week and in the egg yolk at the 11-week after the first immunization.en
dc.description.abstractPenelitian ini bertujuan untuk mempelajari metode produksi antibodi antiekskretori/ sekretori (E/S) Fasciola gigantica pada ayam petelur. 10 ekor ayam dibagi atas 2 kelompok, setiap kelompok terdiri dari 5 ekor ayam. Kelompok pertama diimunisasikan dengan antigen E/S Fasciola gigantica asal kerbau dan kelompok ke-2 diimunisasikan dengan antigen E/S Fasciola gigantica asal domba. Dosis imunisasi tersebut sebanyak 150 μg/ekor. Minggu pertama imunisasi dilakukan melalui intravena tanpa adjuvant. Satu minggu setelahnya (minggu kedua), semua ayam diinjeksikan antigen kombinasi dengan adjuvant komplet dengan perbandingan 1:1 melalui subkutan. Minggu ke-3 hingga minggu ke-6 semua ayam diimunisasi dengan mengkombinasikan antigen dengan adjuvant inkomplet. Telur di koleksi setiap hari. Serum dan kuning telur di uji agar gel presipitation (AGPT) untuk mendeteksi ada atau tidaknya antibodi yang terbentuk. Hasil AGPT menunjukkan bahwa ayam yang diimunisasi dengan antigen asal kerbau lebih cepat dibandingkan dengan ayam yang diimunisasi dengan antigen asal domba. Ayam yang diimunisasikan dengan dengan E/S F.gigantica asal kerbau, antibodi pada serum dan kuning telur ditemukan berturutturut pada minggu ke-9 dan ke-10 setelah imunisasi pertama. Berbeda dengan ayam yang diimunisasikan dengan dengan E/S F.gigantica asal domba, antibodi pada serum dan kunig telur ditemukan berturut-turut pada minggu ke-10 dan ke- 11 setelah imunisasi pertama.
