dc.description.abstractThe study was designed to develop ELISA method to detect coproantigen of Fasciola gigantica in ruminant as a model for human fasciolosis diagnostic test. Fasciolosis Diagnostic Test. Under direction of FADJAR SATRIJA and SRI MURTINI. Excretory Secretory (ES) antigen were isolated from adult stage of F. gigantica incubated in the RPMI medium at 370C for 4 hours. Each worm produced roughly 0.18 mg ES antigen. ES protein showed five protein bands with the molecular weight of 104.99, 90.05, 56.43, 47, 30.27 kDa in SDS-PAGE analysis. P olyclonal antibodies against ES antigen were raised from rabbits and chicken egg yolk. These polyclonal antibodies were used in sandwich ELISA for detection of coproantigen using IgY as capture antibody and IgG as detection antibody. Specificity and sensitivity test revealed that the ELISA have 87,5% sensitivity and 100% specificity compared to gold standard post-mortem examination.en
