variasi parameter. Increased free radical levels in the body can be caused by rising air pollution resulting from industrial development. This condition leads to oxidative stress, which triggers macromolecular damage. Free radicals in the body can be neutralized by antioxidant compounds. Indonesia has a rich biodiversity with the potential to be developed as a source of natural antioxidants. Andaliman fruit (Zanthoxylum acanthopodium DC.) is an indigenous Indonesian plant known to contain polyphenolic compounds with antioxidant potential; however, its utilization remains limited to traditional medicine and culinary seasoning. The quality and quantity of compounds recovered are determined by various factors, including extraction time, solid-to-liquid ratio, and solvent concentration. This study aimed to determine the optimal extraction conditions, based on the combination of extraction time, solid-to-liquid ratio, and ethanol concentration, to maximize the polyphenol content and antioxidant capacity of andaliman fruit using Response Surface Methodology (RSM) with an I-optimal design.
The research method consisted of several stages. The first stage involved optimizing extraction conditions based on an RSM experimental design using an I-optimal design. The optimized extracts were evaluated for total phenolic content (TPC) using the Folin–Ciocalteu method, total flavonoid content (TFC) using the aluminum chloride complexation method, and antioxidant capacity using the DPPH and FRAP assays. The optimal conditions obtained were verified through actual experiments. The subsequent stage compared the TPC and TFC values obtained under the optimal conditions of this study with those reported in previous studies.
The numerical optimization results showed that the optimal formula was obtained with a desirability value of 0.776, consisting of an extraction time of 3 min, a solid-to-liquid ratio of 1:20 g/mL, and an ethanol concentration of 30%. The significance of the model obtained from the combination of extraction parameters was analyzed at a 95% confidence level, and all responses had p-values <0.05. The highest TPC and TFC values were obtained in Run 18, at 45.444 mg GAE/g DW and 2.119 mg GAE/g DW, respectively. Meanwhile, the highest antioxidant capacities by the DPPH and FRAP assays were obtained in Run 2 and Run 20, at 0.882 µmol TE/g DW and 19.333 µmol TE/g DW, respectively. The optimum conditions were an extraction time of 3 min, a solid-to-liquid ratio of 1:20, and an ethanol concentration of 30%, yielding 45.444 mg GAE/g DW and 2.119 mg GAE/g DW. Further identification of the compound profile of andaliman fruit extract is required to determine changes in compound composition resulting from variations in extraction parameters.
