      Abstract
      Tanaman ketepeng cina (Cassia alata L.) mengandung antrakuinon yang berpotensi memberikan efek laksatif dengan menghambat penyerapan air dan meningkatkan motilitas usus. Penelitian ini mengevaluasi efek ekstrak etanol daun ketepeng cina (EKC) terhadap struktur histologis kolon tikus jantan Sprague Dawley pada kondisi fisiologis normal. Tikus diberikan EKC dosis 50, 100, dan 200 mg/kg BB serta bisacodyl sebagai kontrol positif. Preparat histologis sampel kolon dari setiap kelompok diwarnai dengan Alcian Blue (AB) untuk mewarnai musin asam dan Periodic acid-Schiff (PAS) untuk mewarnai musin netral. Parameter yang diamati adalah jumlah, luas area, dan intensitas pewarnaan sel goblet kolon. Hasil menunjukkan pemberian EKC menyebabkan peningkatan signifikan pada seluruh parameter dibandingkan kontrol normal, sesuai dengan mekanisme laksatif. Dosis 50 mg/kg BB menunjukkan profil histologis yang paling mirip dengan kontrol positif dan direkomendasikan sebagai dosis laksatif herbal yang efektif.
       
      Candle bush plants (Cassia alata L.) contains anthraquinones that potentially exert laxative effects by inhibiting water absorption and increasing intestinal motility. This study evaluates the effect of ethanol extract of candle bush leaves (EKC) on the histological structure of the colon in male Sprague Dawley rats under normal physiological conditions. Rats were administered EKC at doses of 50, 100, and 200 mg/kg BW with bisacodyl as a positive control. Histological preparations of rat colon samples from each group were stained with Alcian Blue (AB) to stain acidic mucin and Periodic acid-Schiff (PAS) to stain neutral mucin. The observed parameters were the number, surface area, and staining intensity of colonic goblet cells. Result showed administration of EKC led to a significant increase in all parameters compared to the normal control, consistent with laxative mechanisms. The 50 mg/kg BW dose exhibited a histological profile most similar to the positive control and recommended as the effective herbal laxative dose.
