Perubahan iklim global meningkatkan frekuensi kekeringan, menurunkan ketersediaan air tanah, serta mengancam pertumbuhan bibit tanaman hutan, terutama pada fase awal pertumbuhan, sehingga berpotensi menurunkan produktivitas bahkan menyebabkan kematian tanaman. Meskipun tanaman memiliki mekanisme adaptasi morfofisiologis dan biokimia terhadap cekaman kekeringan, kemampuan tersebut sering kali belum memadai untuk menghadapi kondisi lingkungan yang semakin ekstrem. Salah satu pendekatan bioprospeksi yang berpotensi meningkatkan ketahanan tanaman terhadap kekeringan adalah pemanfaatan dark septate endophytes (DSE), yaitu cendawan endofit berhifa gelap yang membentuk mikrosklerotia pada jaringan akar. DSE diketahui mampu meningkatkan toleransi tanaman terhadap cekaman abiotik, memperbaiki pertumbuhan, dan meningkatkan efisiensi serapan hara. Namun, eksplorasi DSE pada tanaman hutan tropis Indonesia masih terbatas.
Penelitian ini bertujuan mengeksplorasi dan mengkarakterisasi DSE yang berasosiasi dengan akar Neolamarckia cadamba, serta mengevaluasi potensi fungsionalnya dalam meningkatkan pertumbuhan dan ketahanan bibit terhadap cekaman kekeringan. Penelitian terbagi menjadi empat bagian, yaitu (1) eksplorasi dan isolasi DSE dari akar N. cadamba di Bogor, Garut, dan Kediri; (2) karakterisasi morfologi dan molekuler; (3) pengujian kemampuan fungsional; serta (4) evaluasi pengaruh inokulasi DSE terhadap pertumbuhan dan respons bibit dalam kondisi cekaman kekeringan.
Hasil eksplorasi menunjukkan bahwa akar N. cadamba terkolonisasi secara alami oleh DSE dengan tingkat kolonisasi sebesar 65% (Bogor), 40% (Garut), dan 32% (Kediri). Keberadaan DSE diduga berkaitan dengan kondisi tanah yang masam hingga sangat masam (pH 4,4-4,9). Sebanyak 12 isolat cendawan berkoloni gelap berhasil diisolasi, dan berdasarkan karakterisasi morfologi, pengujian biologis menggunakan tanaman indikator, serta evaluasi karakter fisiologis, enam isolat teridentifikasi sebagai kandidat DSE yang cukup prospektif, yaitu CPGS-1, CPGS-3, CPPS-9, CKKS-3, CKKS-5, dan CGKR-9. Keenam isolat tersebut mampu menghasilkan hormon indole-3-acetic acid serta menunjukkan tingkat toleransi yang berbeda terhadap cekaman osmotik PEG 6000, yang mencerminkan keragaman fisiologis dan potensi pembentukan simbiosis dengan tanaman.
Identifikasi molekuler berdasarkan sekuens daerah ITS DNA ribosom berhasil mengungkap identitas taksonomi dan hubungan kekerabatan keenam isolat terpilih. Analisis filogenetik mengelompokkan isolat ke dalam dua kelas utama, yaitu Dothideomycetes dan Eurotiomycetes. Isolat CPGS-1 dan CPPS-9 berkerabat dengan kelompok Dothideomycetes sp.; CGKR-9 berkerabat dengan Exophiala spinifera, sedangkan isolat CKKS-5, CKKS-3, dan CPGS-3 berkerabat dengan genus Knufia. Hasil tersebut menunjukkan bahwa kandidat DSE yang berasosiasi dengan N. cadamba memiliki keragaman genetik dan taksonomi yang tinggi, serta berpotensi dikembangkan sebagai agen hayati untuk mendukung pembangunan hutan tanaman tropis.
Evaluasi efektivitas kandidat DSE pada bibit N. cadamba menunjukkan bahwa peningkatan karakter morfofisiologi bibit sangat dipengaruhi oleh kompatibilitas antara isolat DSE dan provenans tanaman. Pada provenans Kampar, kombinasi provenans Kampar–KSP menghasilkan pertumbuhan terbaik, sedangkan pada provenans Pomalaa, kombinasi Pomalaa–CKKS-5 dan Pomalaa–CPGS-3 menunjukkan efektivitas tertinggi. Hasil tersebut menunjukkan bahwa keberhasilan simbiosis tidak hanya ditentukan oleh karakter fisiologis isolat, tetapi juga oleh variasi genetik tanaman inang.
Pada kondisi cekaman kekeringan, respons bibit dipengaruhi oleh interaksi antara tingkat ketersediaan air dan inokulasi DSE. Isolat CKKS-5 merupakan isolat paling efektif dalam meningkatkan toleransi kekeringan, terutama pada cekaman sedang (50% kapasitas lapang), melalui pemeliharaan pertumbuhan, biomassa, dan fungsi fisiologis yang tercermin dari nilai indeks sensitivitas cekaman (ISC) terendah. Sebaliknya, isolat KSP lebih efektif pada cekaman berat (25% kapasitas lapang) dengan mempertahankan produktivitas bibit melalui kombinasi mekanisme drought avoidance dan drought tolerance, sedangkan CPGS-3 belum mampu menginduksi respons adaptif yang mendukung toleransi kekeringan meskipun tingkat kolonisasi akarnya tinggi.
Penelitian ini menunjukkan bahwa efektivitas DSE dalam meningkatkan adaptasi morfofisiologis bibit N. cadamba terhadap cekaman kekeringan berkaitan dengan karakter fisiologis isolat DSE, variasi genetik provenans tanaman, dan tingkat cekaman yang dihadapi. Evaluasi pada tiga provenans menunjukkan adanya perbedaan respons tanaman terhadap isolat DSE, sedangkan pengujian cekaman kekeringan menunjukkan bahwa efektivitas DSE bersifat isolate-specific dan stress-dependent. Tingkat kolonisasi akar tidak selalu mencerminkan efektivitas DSE dalam meningkatkan adaptasi tanaman terhadap kekeringan. Kebaruan penelitian ini meliputi: (1) keberhasilan mengidentifikasi enam isolat DSE yang berasosiasi dengan akar N. cadamba berdasarkan karakter morfologi dan molekuler, dengan Knufia sp. CKKS-5 sebagai salah satu isolat potensial yang mampu menghasilkan IAA dan mendukung adaptasi bibit pada cekaman kekeringan hingga 50% kapasitas lapang; (2) pengembangan konsep evaluasi efektivitas DSE secara integratif melalui keterkaitan antara karakter fisiologis isolat, variasi provenans tanaman, tingkat kolonisasi akar, dan respons fungsional tanaman; serta (3) pengembangan pendekatan metodologis integratif untuk menyeleksi DSE toleran kekeringan, mulai dari eksplorasi dan karakterisasi in vitro hingga pengujian efektivitasnya secara in vivo pada berbagai tingkat cekaman kekeringan. Global climate change is increasing the frequency and severity of drought events, reducing soil water availability, and threatening the establishment and growth of forest seedlings, particularly during the early stages of development. These conditions may reduce productivity and ultimately lead to seedling mortality. Although plants possess physiological, biochemical, and morphological mechanisms to adapt to drought stress, these mechanisms are often insufficient to cope with increasingly severe environmental conditions. One promising bioprospecting approach to enhance drought tolerance is the utilization of dark septate endophytes (DSE), a group of root-associated endophytic fungi characterized by melanized septate hyphae and microsclerotia. DSE’s have been reported to improve plant growth, nutrient acquisition, and tolerance to abiotic stresses. However, information regarding DSE associated with tropical forest tree species in Indonesia remains limited.
This study aimed to explore and characterize DSE associated with the roots of Neolamarckia cadamba and to evaluate their functional potential in enhancing seedling growth and drought tolerance. The study consisted of four stages: (1) exploration and isolation of DSE from N. cadamba roots collected in Bogor, Garut, and Kediri; (2) morphological and molecular characterization; (3) evaluation of functional traits; and (4) assessment of the effects of DSE inoculation on seedling growth and responses under drought stress.
The exploration revealed that N. cadamba roots were naturally colonized by DSE, with colonization rates of 65% in Bogor, 40% in Garut, and 32% in Kediri. The occurrence of DSE was associated with acidic to strongly acidic soils (pH 4.4–4.9). A total of 12 dark-colony fungal isolates were successfully isolated, of which six isolates (CPGS-1, CPGS-3, CPPS-9, CKKS-3, CKKS-5, and CGKR-9) were identified as DSE candidates based on morphological characteristics, biological assays using indicator plants, and physiological evaluations. All six isolates produced indole-3-acetic acid (IAA) and exhibited different levels of tolerance to PEG 6000-induced osmotic stress, indicating physiological diversity and potential for establishing beneficial symbioses with plants.
Molecular identification based on the internal transcribed spacer (ITS) region of ribosomal DNA successfully determined the taxonomic identity and phylogenetic relationships of the six selected isolates. Phylogenetic analysis classified the isolates into two major fungal classes, Dothideomycetes and Eurotiomycetes. Isolates CPGS-1 and CPPS-9 were affiliated with Dothideomycetes; CGKR-9 showed close similarity to Exophiala spinifera, whereas CKKS-5, CKKS-3, and CPGS-3 were closely related to the genus Knufia. These findings demonstrate the high taxonomic and genetic diversity of DSE associated with N. cadamba and highlight their potential as biological resources for tropical forest plantation development.
Evaluation of the effectiveness of DSE isolates on N. cadamba seedlings demonstrated that improvements in seedling morphophysiological characteristics were strongly influenced by the compatibility between DSE isolates and plant provenances. In the Kampar provenance, the Kampar–KSP combination produced the greatest growth response, whereas in the Pomalaa provenance, the Pomalaa–CKKS-5 and Pomalaa–CPGS-3 combinations exhibited the highest effectiveness. These findings indicate that the success of the symbiotic association is determined not only by the physiological characteristics of the DSE isolates but also by the genetic variation of the host plant.
Under drought stress conditions, seedling responses were influenced by the interaction between water availability and DSE inoculation. The CKKS-5 isolate was the most effective in enhancing drought tolerance, particularly under moderate drought stress (50% field capacity), by maintaining growth, biomass accumulation, and physiological performance, as reflected by the lowest Stress Susceptibility Index (SSI). In contrast, the KSP isolate performed better under severe drought stress (25% field capacity) by sustaining seedling productivity through a combination of drought avoidance and drought tolerance mechanisms. Meanwhile, CPGS-3 failed to induce adaptive responses that effectively enhanced drought tolerance, despite exhibiting a high level of root colonization.
This study demonstrates that the effectiveness of DSE in enhancing the morphophysiological adaptation of N. cadamba seedlings to drought stress is associated with the physiological characteristics of the isolates, their compatibility with the host plant, and the severity of the stress imposed. Evaluation across three provenances revealed differences in plant responses to DSE isolates, whereas the drought-stress experiment demonstrated that DSE effectiveness was isolate-specific and stress-dependent. Root colonization levels did not necessarily reflect the effectiveness of DSE in enhancing plant adaptation to drought stress. The novelty of this research lies in three main aspects: (1) the identification of six DSE isolates associated with the roots of N. cadamba based on morphological and molecular characteristics, including Knufia sp. CKKS-5, a promising isolate capable of producing indole-3-acetic acid (IAA) and supporting seedling adaptation to drought stress at 50% field capacity; (2) the development of an integrative framework for evaluating DSE effectiveness by linking isolate physiological traits, plant provenance variation, root colonization levels, and plant functional responses; and (3) the development of an integrative methodological approach for selecting drought-tolerant DSE, encompassing exploration and in vitro characterization through to in vivo effectiveness testing under different levels of drought stress.
