dc.description.abstractMUHAMMAD FEBRIAN AR RAHMAN. Efek Preventif Ekstrak Lengkuas (Alpinia galanga) terhadap Aorta dan Pankreas Tikus dengan Pakan Tinggi Lemak. Dibimbing oleh EVA HARLINA, GUNANTI, dan HASIM. Pemberian pakan tinggi lemak (PTL) dapat berkaitan dengan perubahan struktural dan respons jaringan pada berbagai organ, termasuk pankreas dan aorta. Penelitian ini bertujuan mengevaluasi efek pemberian preventif ekstrak lengkuas (Alpinia galanga) terhadap profil hemogram, histopatologi pankreas dan aorta, luas pulau Langerhans, ketebalan intima-media aorta, serta imunoreaktivitas insulin pankreas dan TNF-a aorta pada tikus yang diberi PTL. Sebanyak 20 ekor tikus jantan Sprague-Dawley dibagi menjadi empat kelompok (n = 5), yaitu kontrol (KK), kontrol PTL (KN), terapi Orlistat® (KP), dan preventif ekstrak (PE). KK diberi pakan standar selama 10 minggu, sedangkan KN diberi PTL selama 10 minggu. KP diberi PTL selama 10 minggu dengan penambahan Orlistat® 12,5 mg/kgBB/hari pada minggu ke-7-10. PE diberi PTL dan ekstrak A. galanga 500 mg/kgBB/hari secara bersamaan sejak awal hingga akhir periode 10 minggu. Parameter yang dievaluasi meliputi profil hemogram, histopatologi dan morfometri pankreas dan aorta, serta imunoreaktivitas insulin pankreas dan TNF-a aorta. Profil hemogram tidak menunjukkan perbedaan signifikan antarkelompok. Pada pankreas, PE menunjukkan skor kongesti yang lebih rendah dibandingkan KN dan KP serta skor edema yang lebih rendah dibandingkan KN, sedangkan infiltrasi sel radang tidak berbeda signifikan antarkelompok. Luas pulau Langerhans berbeda signifikan antarkelompok, dengan nilai tertinggi pada KN, diikuti KP, PE, dan KK. Imunoreaktivitas insulin pada KN dan KP lebih tinggi dibandingkan KK dan PE, sedangkan PE tidak berbeda signifikan dengan KK. Pada aorta, skor penebalan tunika media pada KN dan KP lebih tinggi dibandingkan KK, sedangkan PE tidak berbeda signifikan dengan kelompok lainnya. Disorganisasi serabut elastis dan infiltrasi sel radang tidak berbeda signifikan antarkelompok. Ketebalan intimamedia pada KN lebih tinggi dibandingkan KK dan PE, sedangkan PE tidak berbeda signifikan dengan KK dan KP. Imunoreaktivitas TNF-a tertinggi ditemukan pada KN, diikuti KP, sedangkan PE tidak berbeda signifikan dengan KK. Secara keseluruhan, pemberian ekstrak A. galanga sejak awal paparan PTL berkaitan dengan lebih rendahnya beberapa perubahan histopatologis, morfometrik, dan imunohistokimia pada pankreas dan aorta dibandingkan kelompok PTL tanpa ekstrak. Temuan ini menunjukkan potensi preventif A. galanga terhadap beberapa perubahan jaringan yang berkaitan dengan paparan PTL, tetapi mekanisme metabolik, inflamasi, dan molekuler yang mendasarinya belum dapat ditetapkan karena tidak diukur secara langsung dalam penelitian ini.

Supervised by EVA HARLINA, GUNANTI, and HASIM. A high-fat diet (HFD) may be associated with structural alterations and tissue responses in various organs, including the pancreas and aorta. This study aimed to evaluate the effects of preventive administration of galangal (Alpinia galanga) extract on the hematological profile, pancreatic and aortic histopathology, Langerhans islet area, aortic intima-media thickness, pancreatic insulin immunoreactivity, and aortic TNF-a immunoreactivity in rats fed an HFD. Twenty male Sprague-Dawley rats were allocated into four groups (n = 5): control (KK), HFD control (KN), Orlistat® treatment (KP), and preventive extract (PE). The KK group received a standard diet for 10 weeks, whereas the KN group received an HFD for 10 weeks. The KP group received an HFD for 10 weeks, with Orlistat® at 12.5 mg/kg BW/day administered during weeks 7–10. The PE group received an HFD concurrently with A. galanga extract at 500 mg/kg BW/day from the beginning to the end of the 10-week experimental period. The evaluated parameters included the hematological profile, pancreatic and aortic histopathology and morphometry, pancreatic insulin immunoreactivity, and aortic TNF-a immunoreactivity. The hematological profile did not differ significantly among groups. In the pancreas, PE showed a lower congestion score than KN and KP and a lower edema score than KN, whereas inflammatory cell infiltration did not differ significantly among groups. Langerhans islet area differed significantly among groups, with the highest value in KN, followed by KP, PE, and KK. Insulin immunoreactivity was higher in KN and KP than in KK and PE, whereas PE did not differ significantly from KK. In the aorta, tunica media thickening scores were higher in KN and KP than in KK, whereas PE did not differ significantly from the other groups. Elastic fiber disorganization and inflammatory cell infiltration did not differ significantly among groups. Aortic intima-media thickness was greater in KN than in KK and PE, whereas PE did not differ significantly from KK and KP. TNF-a immunoreactivity was highest in KN, followed by KP, whereas PE did not differ significantly from KK. Overall, administration of A. galanga extract from the beginning of HFD exposure was associated with lower levels of several histopathological, morphometric, and immunohistochemical alterations in the pancreas and aorta compared with the HFD group without extract administration. These findings indicate the preventive potential of A. galanga against several tissue alterations associated with HFD exposure; however, the underlying metabolic, inflammatory, and molecular mechanisms cannot be established because they were not directly evaluated in this study.
